Sec is encoded by the UGA codon, which normally signals translation termination. The recoding of UGA as Sec codon requires a Sec insertion sequence (SECIS) element; present in the 3' untranslated regions of eukaryotic selenoprotein mRNAs. This protein specifically binds to the SECIS element, which is stimulated by a Sec-specific translation elongation factor. Mutations in this gene have been associated with reduction in enzymatic activity of type II iodothyronine deiodinase (a selenoprotein) and abnormal thyroid hormone metabolism. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Aug 2017]. * **UniProt Summary**: Binds to the SECIS element in the 3'-UTR of some mRNAs encoding selenoproteins. Binding is stimulated by SELB. |Ribosomal L7Ae| |negative regulation of nuclear-transcribed mRNA catabolic process, nonsense-mediated decay| |selenocysteine insertion sequence binding| |regulation of nuclear-transcribed mRNA catabolic process, nonsense-mediated decay| |selenocysteine incorporation| |translational readthrough| |striatum development| |subpallium development| |ribonucleoprotein complex binding| |negative regulation of mRNA catabolic process| |negative regulation of RNA catabolic process| |mRNA 3-UTR binding| |negative regulation of mRNA metabolic process| |translational elongation| |ribonucleoprotein complex| |regulation of mRNA catabolic process| |negative regulation of cellular catabolic process| |telencephalon development| |negative regulation of catabolic process| |regulation of mRNA metabolic process| |regulation of translation| |translation| |forebrain development| |regulation of cellular amide metabolic process| |peptide biosynthetic process| |posttranscriptional regulation of gene expression| |amide biosynthetic process| |peptide metabolic process| |brain development| |head development| |cellular amide metabolic process| |neuron development| |regulation of cellular catabolic process| |central nervous system development| |regulation of catabolic process| |neuron differentiation| |mitochondrion| |negative regulation of RNA metabolic process| |RNA binding| |organonitrogen compound biosynthetic process| |negative regulation of nucleobase-containing compound metabolic process| |generation of neurons| |neurogenesis| |cellular nitrogen compound biosynthetic process| |cell development| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |positive regulation of gene expression| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Radicicol 0.16μM R05 exp242|-2.19| |Artemisinin 50μM R08 exp450|-2.13| |Senexin-A 25μM R07 exp400|-1.99| |Palbociclib 1μM R07 exp382|-1.94| |Vinblastine 0.015μM R05 exp208|-1.94| |UM0130462 0.025 to 0.035μM day4 R05 exp247|-1.93| |MLN-4924 2μM R00 exp22|-1.89| |5-Fluorouracil 20μM R00 exp2|-1.88| |Lead acetate 2000μM R08 exp498 no dilution day6|-1.83| |GSK461364A 0.1μM R03 exp126|-1.79| |Trientine 500μM R08 exp534|-1.76| |Q15 2.7μM R02 exp79|1.72| |MK-1775 0.32μM R02 exp75|1.97| ^Gene^Correlation^ |PSTK|0.421| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 2421 * **Expression level (log2 read counts)**: 6.17
