This inhibition is most likely accomplished by masking histone lysines from being acetylated, and the consequence is to silence HAT-dependent transcription. The encoded protein is part of a complex localized to the endoplasmic reticulum but is found in the nucleus and inhibits apoptosis following attack by cytotoxic T lymphocytes. This protein can also enhance DNA replication of the adenovirus genome. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]. * **UniProt Summary**: Multitasking protein, involved in apoptosis, transcription, nucleosome assembly and histone chaperoning. Isoform 2 anti-apoptotic activity is mediated by inhibition of the GZMA-activated DNase, NME1. In the course of cytotoxic T- lymphocyte (CTL)-induced apoptosis, GZMA cleaves SET, disrupting its binding to NME1 and releasing NME1 inhibition. Isoform 1 and isoform 2 are potent inhibitors of protein phosphatase 2A. Isoform 1 and isoform 2 inhibit EP300/CREBBP and PCAF-mediated acetylation of histones (HAT) and nucleosomes, most probably by masking the accessibility of lysines of histones to the acetylases. The predominant target for inhibition is histone H4. HAT inhibition leads to silencing of HAT-dependent transcription and prevents active demethylation of DNA. Both isoforms stimulate DNA replication of the adenovirus genome complexed with viral core proteins; however, isoform 2 specific activity is higher. {ECO:0000269|PubMed:11555662, ECO:0000269|PubMed:12628186}. |NAP| |negative regulation of histone acetylation| |negative regulation of peptidyl-lysine acetylation| |nucleosome disassembly| |chromatin disassembly| |protein-DNA complex disassembly| |negative regulation of protein acetylation| |protein phosphatase regulator activity| |protein phosphatase inhibitor activity| |negative regulation of histone modification| |regulation of histone acetylation| |negative regulation of phosphoprotein phosphatase activity| |negative regulation of chromatin organization| |regulation of peptidyl-lysine acetylation| |negative regulation of protein dephosphorylation| |regulation of protein acetylation| |lipid droplet| |negative regulation of phosphatase activity| |negative regulation of dephosphorylation| |nucleosome assembly| |regulation of phosphoprotein phosphatase activity| |histone binding| |negative regulation of chromosome organization| |chromatin assembly| |regulation of protein dephosphorylation| |negative regulation of neuron apoptotic process| |regulation of histone modification| |chromatin assembly or disassembly| |nucleosome organization| |regulation of phosphatase activity| |DNA packaging| |regulation of mRNA stability| |regulation of RNA stability| |regulation of chromatin organization| |protein-DNA complex assembly| |regulation of mRNA catabolic process| |negative regulation of neuron death| |regulation of neuron apoptotic process| |regulation of dephosphorylation| |DNA replication| |protein-containing complex disassembly| |protein-DNA complex subunit organization| |DNA conformation change| |regulation of neuron death| |regulation of mRNA metabolic process| |regulation of chromosome organization| |negative regulation of organelle organization| |cellular component disassembly| |negative regulation of hydrolase activity| |posttranscriptional regulation of gene expression| |negative regulation of phosphate metabolic process| |negative regulation of phosphorus metabolic process| |negative regulation of protein modification process| |protein-containing complex| |perinuclear region of cytoplasm| |chromatin organization| |viral process| |negative regulation of cellular component organization| |symbiotic process| |negative regulation of catalytic activity| |interspecies interaction between organisms| |regulation of cellular catabolic process| |cellular protein-containing complex assembly| |negative regulation of apoptotic process| |negative regulation of programmed cell death| |negative regulation of cell death| |regulation of catabolic process| |endoplasmic reticulum| |negative regulation of cellular protein metabolic process| |chromosome organization| |negative regulation of protein metabolic process| |negative regulation of molecular function| |negative regulation of transcription, DNA-templated| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |regulation of hydrolase activity| |regulation of organelle organization| |negative regulation of RNA metabolic process| |negative regulation of cellular macromolecule biosynthetic process| |negative regulation of nucleobase-containing compound metabolic process| |DNA binding| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |regulation of apoptotic process| |negative regulation of biosynthetic process| |protein-containing complex assembly| |regulation of programmed cell death| |regulation of cell death| |cellular macromolecule biosynthetic process| |negative regulation of gene expression| |macromolecule biosynthetic process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| |protein-containing complex subunit organization| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp66|BI-D1870 3.15μM R02 exp66]]|-1.87| |[[:results:exp184|Ixabepilone 0.004 to 0.005μM on day4 R04 exp184]]|-1.82| No correlation found to any other genes in chemogenomics.
