Does not act on ceramide 1- phoshate, lysophosphatidic acid or phosphatidic acid. Regulates the intracellular levels of the bioactive sphingolipid metabolite S1P that regulates diverse biological processes acting both as an extracellular receptor ligand or as an intracellular second messenger (PubMed:12815058, PubMed:11756451). Involved in efficient ceramide synthesis from exogenous sphingoid bases. Converts S1P to sphingosine, which is readily metabolized to ceramide via ceramide synthase. In concert with sphingosine kinase 2 (SphK2), recycles sphingosine into ceramide though a phosphorylation/dephosphorylation cycle. Regulates intracellular ceramide levels, which in turn regulate apoptosis (By similarity). Via S1P levels, modulates resting tone, intracellular Ca(2+) and myogenic vasoconstriction in resistance arteries (PubMed:18583713). Also involved in unfolded protein response (UPR) and ER stress-induced autophagy via regulation of intracellular S1P levels (PubMed:20798685). {ECO:0000250|UniProtKB:Q9JI99, ECO:0000269|PubMed:11756451, ECO:0000269|PubMed:12815058, ECO:0000269|PubMed:18583713, ECO:0000269|PubMed:20798685}. |PAP2| |sphinganine-1-phosphate metabolic process| |sphingosine-1-phosphate phosphatase activity| |ER to Golgi ceramide transport| |sphingosine metabolic process| |ceramide transport| |sphingoid metabolic process| |diol metabolic process| |intracellular lipid transport| |regulation of keratinocyte differentiation| |regulation of epidermal cell differentiation| |regulation of epidermis development| |sphingolipid biosynthetic process| |extrinsic apoptotic signaling pathway| |polyol metabolic process| |membrane lipid biosynthetic process| |regulation of epithelial cell differentiation| |intrinsic apoptotic signaling pathway| |sphingolipid metabolic process| |ammonium ion metabolic process| |membrane lipid metabolic process| |apoptotic signaling pathway| |lipid transport| |dephosphorylation| |alcohol metabolic process| |lipid localization| |phospholipid metabolic process| |organic hydroxy compound metabolic process| |lipid biosynthetic process| |organophosphate metabolic process| |apoptotic process| |endoplasmic reticulum membrane| |cellular lipid metabolic process| |programmed cell death| |cell death| |lipid metabolic process| |organonitrogen compound biosynthetic process| |intracellular transport| |amide transport| |intracellular signal transduction| |small molecule metabolic process| |regulation of cell differentiation| |establishment of localization in cell| |nitrogen compound transport| \\ === CRISPR Data === No hits were found. ^Gene^Correlation^ |[[:human genes:h:hgc6.3|HGC6.3]]|0.616| Global Fraction of Cell Lines Where Essential: 5/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|1/26| |breast|0/33| |central nervous system|1/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|2/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 4920 * **Expression level (log2 read counts)**: 4.14 {{:chemogenomics:nalm6 dist.png?nolink |}}
