Mutations in this gene are associated with erythrocyte lactate transporter defect. Alternatively spliced transcript variants have been found for this gene.[provided by RefSeq, Oct 2009]. * **UniProt Summary**: Proton-coupled monocarboxylate transporter. Catalyzes the rapid transport across the plasma membrane of many monocarboxylates such as lactate, pyruvate, branched-chain oxo acids derived from leucine, valine and isoleucine, and the ketone bodies acetoacetate, beta-hydroxybutyrate and acetate. Depending on the tissue and on cicumstances, mediates the import or export of lactic acid and ketone bodies. Required for normal nutrient assimilation, increase of white adipose tissue and body weight gain when on a high-fat diet. Plays a role in cellular responses to a high-fat diet by modulating the cellular levels of lactate and pyruvate, small molecules that contribute to the regulation of central metabolic pathways and insulin secretion, with concomitant effects on plasma insulin levels and blood glucose homeostasis. {ECO:0000269|PubMed:17701893}. |MFS 1| |mevalonate transport| |mevalonate transmembrane transporter activity| |behavioral response to nutrient| |organic cyclic compound binding| |plasma membrane lactate transport| |lactate transmembrane transporter activity| |lactate transport| |lactate transmembrane transport| |monocarboxylic acid transmembrane transporter activity| |response to food| |symporter activity| |pyruvate metabolic process| |centrosome cycle| |microtubule organizing center organization| |organic acid transmembrane transport| |carboxylic acid transmembrane transport| |monocarboxylic acid transport| |organic hydroxy compound transport| |regulation of insulin secretion| |glucose homeostasis| |carbohydrate homeostasis| |regulation of peptide hormone secretion| |response to nutrient| |regulation of hormone secretion| |synapse| |anion transmembrane transport| |carboxylic acid transport| |organic acid transport| |leukocyte migration| |organic anion transport| |regulation of protein secretion| |microtubule cytoskeleton organization| |centrosome| |regulation of peptide secretion| |response to nutrient levels| |monocarboxylic acid metabolic process| |response to extracellular stimulus| |regulation of hormone levels| |cellular response to organic cyclic compound| |cell junction| |behavior| |anion transport| |microtubule-based process| |intracellular membrane-bounded organelle| |regulation of protein transport| |regulation of peptide transport| |regulation of establishment of protein localization| |regulation of secretion by cell| |regulation of secretion| |protein homodimerization activity| |carboxylic acid metabolic process| |response to organic cyclic compound| |ion transmembrane transport| |cell migration| |cell cycle process| |oxoacid metabolic process| |organic acid metabolic process| |regulation of protein localization| |localization of cell| |cell motility| |cytoskeleton organization| |chemical homeostasis| |lipid metabolic process| |transmembrane transport| |locomotion| |cell cycle| |ion transport| |integral component of plasma membrane| |movement of cell or subcellular component| |homeostatic process| |small molecule metabolic process| |regulation of transport| |membrane| \\ === CRISPR Data === ^Screen^Score^ |Bortezomib 0.05μM R00 exp7|-2.59| |QNZ 0.01μM R05 exp241|-2.39| |Mubritinib 0.2μM R01 exp48|-2.29| |Rotenone 0.07μM R08 exp431|-2.19| |QNZ 0.01μM R08 exp439|-2| |IWR1 50μM R08 exp495|-1.87| |Rotenone 20μM R00 exp34|-1.86| |Nicotinate 1000μM R03 exp140|-1.72| |Nutlin-3A 1.6μM R06 exp294|2.38| |Thapsigargin 0.005μM R07 exp407|2.4| |2-Deoxy-D-glucose 800μM R05 exp214|2.67| |Aminopterin 0.005μM R07 exp335|3.19| |3-Bromopyruvate 7μM R04 exp175|4.17| ^Gene^Correlation^ |BSG|0.562| |PFKP|0.428| Global Fraction of Cell Lines Where Essential: 5/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|1/21| |liver|0/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|1/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 136 * **Expression level (log2 read counts)**: 8.58
