Succinyl-CoA synthetase catalyzes the reversible reaction involving the formation of succinyl-CoA and succinate. Alternate splicing results in multiple transcript variants. Pseudogenes of this gene are found on chromosomes 5 and 12. [provided by RefSeq, Apr 2010]. * **UniProt Summary**: GTP-specific succinyl-CoA synthetase functions in the citric acid cycle (TCA), coupling the hydrolysis of succinyl-CoA to the synthesis of GTP and thus represents the only step of substrate-level phosphorylation in the TCA. The beta subunit provides nucleotide specificity of the enzyme and binds the substrate succinate, while the binding sites for coenzyme A and phosphate are found in the alpha subunit. {ECO:0000255|HAMAP- Rule:MF_03221}. |ATP-grasp 2| |Ligase CoA| |succinate-CoA ligase complex (GDP-forming)| |succinate-CoA ligase complex| |succinate-CoA ligase (GDP-forming) activity| |succinate-CoA ligase (ADP-forming) activity| |succinyl-CoA metabolic process| |succinate metabolic process| |tricarboxylic acid cycle| |GDP binding| |aerobic respiration| |thioester metabolic process| |dicarboxylic acid metabolic process| |acyl-CoA metabolic process| |ribonucleoside bisphosphate metabolic process| |purine nucleoside bisphosphate metabolic process| |nucleoside bisphosphate metabolic process| |cellular respiration| |magnesium ion binding| |energy derivation by oxidation of organic compounds| |coenzyme metabolic process| |purine ribonucleotide metabolic process| |ribonucleotide metabolic process| |purine nucleotide metabolic process| |ribose phosphate metabolic process| |sulfur compound metabolic process| |purine-containing compound metabolic process| |mitochondrial matrix| |GTP binding| |generation of precursor metabolites and energy| |cofactor metabolic process| |nucleotide metabolic process| |nucleoside phosphate metabolic process| |protein heterodimerization activity| |drug metabolic process| |nucleobase-containing small molecule metabolic process| |cellular amide metabolic process| |organophosphate metabolic process| |carboxylic acid metabolic process| |oxidation-reduction process| |oxoacid metabolic process| |organic acid metabolic process| |carbohydrate derivative metabolic process| |mitochondrion| |ATP binding| |small molecule metabolic process| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp501|Methotrexate 0.01μM R08 exp501]]|-1.84| No correlation found to any other genes in chemogenomics.
