The encoded protein promotes the expression of genes downstream of transforming growth factor beta (TGF-beta) signaling. This gene may be involved in chromosomal translocations in renal cell carcinomas and other cancers, resulting in the production of fusion proteins. Translocation partners include PRCC (papillary renal cell carcinoma), NONO (non-POU domain containing, octamer-binding), and ASPSCR1 (alveolar soft part sarcoma chromosome region, candidate 1), among other genes. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Aug 2013]. * **UniProt Summary**: Transcription factor that specifically recognizes and binds E-box sequences (5'-CANNTG-3'). Efficient DNA-binding requires dimerization with itself or with another MiT/TFE family member such as TFEB or MITF. In association with TFEB, activates the expression of CD40L in T-cells, thereby playing a role in T- cell-dependent antibody responses in activated CD4(+) T-cells and thymus-dependent humoral immunity. Specifically recognizes the MUE3 box, a subset of E-boxes, present in the immunoglobulin enhancer. It also binds very well to a USF/MLTF site. |DUF3371| |HLH| |negative regulation of cold-induced thermogenesis| |regulation of osteoclast differentiation| |regulation of myeloid leukocyte differentiation| |regulation of cold-induced thermogenesis| |protein dimerization activity| |transcription regulatory region DNA binding| |regulation of myeloid cell differentiation| |regulation of leukocyte differentiation| |humoral immune response| |positive regulation of cell adhesion| |DNA-binding transcription activator activity, RNA polymerase II-specific| |regulation of hemopoiesis| |RNA polymerase II proximal promoter sequence-specific DNA binding| |adaptive immune response| |DNA-binding transcription factor activity| |regulation of cell adhesion| |negative regulation of multicellular organismal process| |positive regulation of transcription by RNA polymerase II| |positive regulation of transcription, DNA-templated| |DNA-binding transcription factor activity, RNA polymerase II-specific| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |regulation of immune system process| |positive regulation of RNA metabolic process| |regulation of cell differentiation| |immune response| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Cerivastatin 0.15μM R05 exp226|-1.85| |BI-D1870 2μM R04 exp162|-1.73| |Dynasore 10μM R04 exp180|1.7| |Pterostilbene 16μM R02 exp78|1.73| No correlation found to any other genes in chemogenomics.
