Efficient DNA-binding requires dimerization with itself or with another MiT/TFE family member such as TFE3 or MITF. In association with TFE3, activates the expression of CD40L in T-cells, thereby playing a role in T- cell-dependent antibody responses in activated CD4(+) T-cells and thymus-dependent humoral immunity. Specifically recognizes and binds the CLEAR-box sequence (5'-GTCACGTGAC-3') present in the regulatory region of many lysosomal genes, leading to activate their expression. It thereby plays a central role in expression of lysosomal genes. Acts as a positive regulator of autophagy by promoting expression of genes involved in autophagy. Specifically recognizes the gamma-E3 box, a subset of E-boxes, present in the heavy-chain immunoglobulin enhancer. Plays a role in the signal transduction processes required for normal vascularization of the placenta. {ECO:0000269|PubMed:19556463, ECO:0000269|PubMed:23434374}. |DUF3371| |HLH| |lysosome localization| |lytic vacuole organization| |lysosome organization| |embryonic placenta development| |positive regulation of autophagy| |vacuole organization| |placenta development| |protein dimerization activity| |transcription regulatory region DNA binding| |autophagy| |process utilizing autophagic mechanism| |regulation of autophagy| |humoral immune response| |positive regulation of cellular catabolic process| |in utero embryonic development| |reproductive structure development| |reproductive system development| |positive regulation of catabolic process| |embryonic organ development| |organelle localization| |adaptive immune response| |chordate embryonic development| |embryo development ending in birth or egg hatching| |developmental process involved in reproduction| |DNA-binding transcription factor activity| |regulation of cellular catabolic process| |embryo development| |regulation of catabolic process| |positive regulation of transcription by RNA polymerase II| |reproductive process| |reproduction| |positive regulation of transcription, DNA-templated| |DNA-binding transcription factor activity, RNA polymerase II-specific| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |positive regulation of RNA metabolic process| |cellular catabolic process| |immune response| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |LB-100 2μM R05 exp210|-1.89| |UM0125461 0.74μM R02 exp84|-1.88| |Vemurafenib 6.6μM R08 exp434|-1.86| |FK-506 30μM R07 exp359|-1.85| |INK128 0.2μM R02 exp70|-1.81| |Colchicine 0.009μM R05 exp215|-1.8| |Pyridostatin 4μM R05 exp240|-1.79| |LB-100 4.1μM R02 exp72|-1.78| |LY2090314 0.003μM R08 exp500 no dilution day6|-1.74| |GSK-J5 1.5μM R08 exp484|-1.73| |Heregulin-B 44ng/ml R08 exp486|-1.73| |BVD-523 15μM R02 exp67|1.89| |Azithromycin 100μM R08 exp452|1.99| |UNC1999 2μM R03 exp155|2.13| No correlation found to any other genes in chemogenomics.
