Isoform 1 inhibits catalytic activity of the PP2A(D) core complex in vitro. The PP2A(C):TIPRL complex does not show phosphatase activity. Acts as negative regulator of serine/threonine-protein phosphatase 4 probably by inhibiting the formation of the active PPP4C:PPP4R2 complex; the function is proposed to implicate it in DNA damage response by promoting H2AFX phosphorylated on Ser-140 (gamma-H2AFX). May play a role in the regulation of ATM/ATR signaling pathway controlling DNA replication and repair. {ECO:0000269|PubMed:17384681, ECO:0000269|PubMed:26717153}. |TIP41| |TOR signaling| |negative regulation of phosphoprotein phosphatase activity| |negative regulation of protein dephosphorylation| |negative regulation of phosphatase activity| |negative regulation of dephosphorylation| |regulation of phosphoprotein phosphatase activity| |DNA damage checkpoint| |regulation of protein dephosphorylation| |DNA integrity checkpoint| |regulation of phosphatase activity| |cell cycle checkpoint| |regulation of dephosphorylation| |negative regulation of hydrolase activity| |negative regulation of phosphate metabolic process| |negative regulation of phosphorus metabolic process| |negative regulation of cell cycle| |negative regulation of protein modification process| |cellular response to DNA damage stimulus| |negative regulation of catalytic activity| |negative regulation of cellular protein metabolic process| |negative regulation of protein metabolic process| |negative regulation of molecular function| |regulation of cell cycle| |regulation of hydrolase activity| |intracellular signal transduction| |cellular response to stress| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| \\ === CRISPR Data === ^Screen^Score^ |Rapamycin 10μM R00 exp30|-2.34| |MK-1775 0.32μM R02 exp75|-2.01| |INK128 0.2μM R02 exp70|-1.98| |Torin1 0.08μM R02 exp82|-1.83| |Senexin-A 25μM R07 exp400|1.8| |Radicicol 0.16μM R05 exp242|1.82| |Artemisinin 50μM R08 exp450|1.83| |Methotrexate 0.01μM R08 exp501|1.85| |2-Methoxyestradiol 0.55μM R07 exp310|2.01| |Arsenate 40μM R06 exp264|2.04| |BML-284 0.09μM R08 exp460|2.05| |VER-155008 3.9μM R06 exp301|2.06| |Palbociclib 1μM R07 exp382|2.1| |Aminopterin 0.005μM R07 exp335|2.12| |Bisphenol S 100μM R08 exp458|2.2| |Thapsigargin 0.005μM R07 exp407|2.36| |Colchicine 0.009μM R05 exp215|2.5| |HMS-I1 1μM R01 exp46|2.57| |Mdivi-1 15μM R05 exp217|2.83| |2-Methoxyestradiol 0.55 to 0.75μM on day4 R07 exp311|2.85| |Milciclib 2μM R08 exp502|3.37| |2-Methoxyestradiol 0.55 to 1μM on day4 R07 exp312|3.65| |UMK57 1μM R01 exp59|4.15| |BVD-523 15μM R02 exp67|4.51| |RO-3307 4.7μM R02 exp80|5.04| ^Gene^Correlation^ |PPP2R4|0.682| |LCMT1|0.604| |PPME1|0.559| |CYB5R4|0.474| |PPP6R3|0.448| |PPP2R1A|0.439| Global Fraction of Cell Lines Where Essential: 20/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|1/26| |breast|1/33| |central nervous system|2/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|1/16| |kidney|1/21| |liver|2/20| |lung|0/75| |lymphocyte|0/16| |ovary|1/26| |pancreas|1/24| |peripheral nervous system|1/16| |plasma cell|0/15| |prostate|0/1| |skin|4/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|2/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1864 * **Expression level (log2 read counts)**: 5.86
