The encoded enzyme localizes to the mitochondria and is required for mitochondrial DNA synthesis. Mutations in this gene are associated with a myopathic form of mitochondrial DNA depletion syndrome. Alternate splicing results in multiple transcript variants encoding distinct isoforms, some of which lack transit peptide, so are not localized to mitochondria. [provided by RefSeq, Dec 2012]. * **UniProt Summary**: Phosphorylates thymidine, deoxycytidine, and deoxyuridine in the mitochondrial matrix. In non-replicating cells, where cytosolic dNTP synthesis is down-regulated, mtDNA synthesis depends solely on TK2 and DGUOK. Widely used as target of antiviral and chemotherapeutic agents. {ECO:0000269|PubMed:11687801}. |dNK| |deoxycytidine metabolic process| |nucleoside kinase activity| |thymidine kinase activity| |deoxycytidine kinase activity| |deoxynucleoside kinase activity| |thymidine metabolic process| |pyrimidine deoxyribonucleoside metabolic process| |deoxyribonucleoside metabolic process| |deoxyribonucleoside monophosphate biosynthetic process| |pyrimidine-containing compound salvage| |pyrimidine nucleoside salvage| |deoxyribonucleoside monophosphate metabolic process| |pyrimidine nucleoside biosynthetic process| |nucleoside salvage| |cellular metabolic compound salvage| |pyrimidine nucleoside metabolic process| |nucleoside biosynthetic process| |glycosyl compound biosynthetic process| |nucleoside monophosphate biosynthetic process| |pyrimidine-containing compound biosynthetic process| |nucleoside monophosphate metabolic process| |nucleobase-containing small molecule biosynthetic process| |pyrimidine-containing compound metabolic process| |nucleoside metabolic process| |DNA biosynthetic process| |glycosyl compound metabolic process| |nucleotide biosynthetic process| |nucleoside phosphate biosynthetic process| |mitochondrial matrix| |nucleotide metabolic process| |nucleoside phosphate metabolic process| |organophosphate biosynthetic process| |nucleobase-containing small molecule metabolic process| |small molecule biosynthetic process| |carbohydrate derivative biosynthetic process| |DNA metabolic process| |organophosphate metabolic process| |carbohydrate derivative metabolic process| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |mitochondrion| |phosphorylation| |organic cyclic compound biosynthetic process| |organonitrogen compound biosynthetic process| |ATP binding| |cellular nitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |small molecule metabolic process| |macromolecule biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp68|Clomiphene 4.4μM R02 exp68]]|-1.99| |[[:results:exp71|KU-0063794 3.8μM R02 exp71]]|-1.73| No correlation found to any other genes in chemogenomics.
