This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine, which exists in both membrane-bound and secreted forms, can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis. Alternative splicing results in multiple transcript variants. Some transcripts skip the last exon of this gene and continue into the second exon of the neighboring TNFSF13 gene; such read-through transcripts are contained in GeneID 407977, TNFSF12-TNFSF13. [provided by RefSeq, Oct 2010]. * **UniProt Summary**: Binds to FN14 and possibly also to TNRFSF12/APO3. Weak inducer of apoptosis in some cell types. Mediates NF-kappa-B activation. Promotes angiogenesis and the proliferation of endothelial cells. Also involved in induction of inflammatory cytokines. Promotes IL8 secretion. {ECO:0000269|PubMed:10085077, ECO:0000269|PubMed:23667509}. |TNF| |tumor necrosis factor receptor binding| |positive regulation of extrinsic apoptotic signaling pathway| |endothelial cell migration| |epithelial cell migration| |epithelium migration| |tissue migration| |positive regulation of endothelial cell proliferation| |extrinsic apoptotic signaling pathway| |tumor necrosis factor-mediated signaling pathway| |regulation of endothelial cell proliferation| |regulation of extrinsic apoptotic signaling pathway| |positive regulation of angiogenesis| |ameboidal-type cell migration| |cytokine activity| |positive regulation of apoptotic signaling pathway| |positive regulation of vasculature development| |positive regulation of epithelial cell proliferation| |positive regulation of protein catabolic process| |cellular response to tumor necrosis factor| |response to tumor necrosis factor| |apoptotic signaling pathway| |regulation of angiogenesis| |angiogenesis| |regulation of vasculature development| |regulation of epithelial cell proliferation| |signaling receptor binding| |regulation of protein catabolic process| |regulation of apoptotic signaling pathway| |blood vessel morphogenesis| |positive regulation of catabolic process| |blood vessel development| |vasculature development| |cardiovascular system development| |positive regulation of apoptotic process| |positive regulation of programmed cell death| |tube morphogenesis| |cytokine-mediated signaling pathway| |positive regulation of cell death| |perinuclear region of cytoplasm| |tube development| |circulatory system development| |anatomical structure formation involved in morphogenesis| |positive regulation of cell population proliferation| |apoptotic process| |cell migration| |regulation of catabolic process| |cellular response to cytokine stimulus| |programmed cell death| |regulation of anatomical structure morphogenesis| |cell motility| |localization of cell| |cell death| |response to cytokine| |locomotion| |positive regulation of developmental process| |integral component of plasma membrane| |regulation of apoptotic process| |movement of cell or subcellular component| |regulation of programmed cell death| |extracellular space| |regulation of cell population proliferation| |positive regulation of signal transduction| |regulation of cell death| |positive regulation of protein metabolic process| |positive regulation of multicellular organismal process| |positive regulation of cell communication| |positive regulation of signaling| |immune response| |extracellular region| \\ === CRISPR Data === ^Screen^Score^ |D-Fructose 10000μM R06 exp279|-2.13| |Cadmium 60μM R08 exp462|-1.87| |A-769662 20μM R07 exp331|1.71| No correlation found to any other genes in chemogenomics.
