Acts as an activator of the Wnt signaling pathway by mediating poly-ADP-ribosylation of AXIN1 and AXIN2, 2 key components of the beta-catenin destruction complex: poly-ADP- ribosylated target proteins are recognized by RNF146, which mediates their ubiquitination and subsequent degradation. Also mediates poly-ADP-ribosylation of BLZF1 and CASC3, followed by recruitment of RNF146 and subsequent ubiquitination. Mediates poly-ADP-ribosylation of TERF1, thereby contributing to the regulation of telomere length. May also regulate vesicle trafficking and modulate the subcellular distribution of SLC2A4/GLUT4-vesicles. Stimulates 26S proteasome activity. {ECO:0000269|PubMed:11739745, ECO:0000269|PubMed:11802774, ECO:0000269|PubMed:19759537, ECO:0000269|PubMed:21478859, ECO:0000269|PubMed:23622245}. |Ank| |Ank 2| |SAM 2| |PARP| |SAM 1| |protein poly-ADP-ribosylation| |protein auto-ADP-ribosylation| |protein localization to chromosome, telomeric region| |protein ADP-ribosylase activity| |positive regulation of telomere capping| |pericentriolar material| |regulation of telomere capping| |negative regulation of telomere maintenance via telomere lengthening| |NAD+ ADP-ribosyltransferase activity| |protein ADP-ribosylation| |positive regulation of telomere maintenance via telomerase| |positive regulation of telomere maintenance via telomere lengthening| |negative regulation of telomere maintenance| |positive regulation of telomere maintenance| |regulation of telomere maintenance via telomerase| |regulation of telomere maintenance via telomere lengthening| |protein localization to chromosome| |regulation of multicellular organism growth| |positive regulation of DNA biosynthetic process| |regulation of telomere maintenance| |multicellular organism growth| |nuclear chromosome, telomeric region| |regulation of DNA biosynthetic process| |negative regulation of DNA metabolic process| |negative regulation of chromosome organization| |positive regulation of canonical Wnt signaling pathway| |positive regulation of chromosome organization| |positive regulation of Wnt signaling pathway| |nuclear envelope| |positive regulation of DNA metabolic process| |regulation of canonical Wnt signaling pathway| |protein polyubiquitination| |regulation of developmental growth| |enzyme binding| |regulation of chromosome organization| |regulation of DNA metabolic process| |Wnt signaling pathway| |cell-cell signaling by wnt| |regulation of Wnt signaling pathway| |negative regulation of organelle organization| |developmental growth| |growth| |cell surface receptor signaling pathway involved in cell-cell signaling| |Golgi membrane| |positive regulation of organelle organization| |regulation of growth| |protein ubiquitination| |perinuclear region of cytoplasm| |negative regulation of cellular component organization| |protein localization to organelle| |protein modification by small protein conjugation| |protein modification by small protein conjugation or removal| |cell-cell signaling| |positive regulation of cellular component organization| |regulation of organelle organization| |negative regulation of nucleobase-containing compound metabolic process| |cellular protein localization| |cellular macromolecule localization| |positive regulation of signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Methotrexate 0.01μM R08 exp501|1.82| |Temozolomide 200μM R04 exp189|1.87| |2-Deoxy-D-glucose 800μM R05 exp214|1.89| |VER-155008 3.9μM R06 exp301|1.96| |LY2090314 0.003μM R08 exp500 no dilution day6|4.14| |LY2090314 0.003μM R08 exp499|4.45| No correlation found to any other genes in chemogenomics.
