The protein contains four ankyrin repeats and an SH3 domain involved in protein-protein interactions. It is localized to the perinuclear region of the cytoplasm, and regulates apoptosis and cell growth through interactions with other regulatory molecules including members of the p53 family. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Regulator that plays a central role in regulation of apoptosis and cell growth via its interactions. Regulates TP53 by enhancing the DNA binding and transactivation function of TP53 on the promoters of proapoptotic genes in vivo. Inhibits the ability of APPBP1 to conjugate NEDD8 to CUL1, and thereby decreases APPBP1 ability to induce apoptosis. Impedes cell cycle progression at G2/M. Its apoptosis-stimulating activity is inhibited by its interaction with DDX42. {ECO:0000269|PubMed:11684014, ECO:0000269|PubMed:12694406, ECO:0000269|PubMed:19377511}. |SH3 2| |SH3 1| |Ank 2| |Ank| |positive regulation of execution phase of apoptosis| |positive regulation of protein insertion into mitochondrial membrane involved in apoptotic signaling pathway| |regulation of protein insertion into mitochondrial membrane involved in apoptotic signaling pathway| |NF-kappaB binding| |positive regulation of mitochondrial outer membrane permeabilization involved in apoptotic signaling pathway| |regulation of execution phase of apoptosis| |regulation of mitochondrial outer membrane permeabilization involved in apoptotic signaling pathway| |intrinsic apoptotic signaling pathway by p53 class mediator| |SH3/SH2 adaptor activity| |positive regulation of establishment of protein localization to mitochondrion| |p53 binding| |regulation of mitochondrial membrane permeability| |regulation of establishment of protein localization to mitochondrion| |regulation of membrane permeability| |positive regulation of neuron death| |positive regulation of mitochondrion organization| |signal transduction by p53 class mediator| |positive regulation of protein localization to membrane| |mitochondrial membrane organization| |SH3 domain binding| |intrinsic apoptotic signaling pathway| |positive regulation of apoptotic signaling pathway| |regulation of signal transduction by p53 class mediator| |regulation of mitochondrion organization| |regulation of protein localization to membrane| |mitochondrial transport| |apoptotic signaling pathway| |regulation of neuron death| |positive regulation of cellular protein localization| |regulation of apoptotic signaling pathway| |mitochondrion organization| |positive regulation of establishment of protein localization| |regulation of cellular protein localization| |cell junction| |negative regulation of cell cycle| |positive regulation of organelle organization| |positive regulation of apoptotic process| |positive regulation of programmed cell death| |perinuclear region of cytoplasm| |positive regulation of cell death| |regulation of establishment of protein localization| |membrane organization| |regulation of cellular localization| |apoptotic process| |positive regulation of transport| |regulation of protein localization| |programmed cell death| |identical protein binding| |cell death| |regulation of cell cycle| |positive regulation of cellular component organization| |mitochondrion| |regulation of organelle organization| |cell cycle| |regulation of apoptotic process| |regulation of programmed cell death| |positive regulation of signal transduction| |regulation of cell death| |intracellular signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |regulation of transport| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp459|Bleomycin 5μM R08 exp459]]|-2.07| |[[:results:exp189|Temozolomide 200μM R04 exp189]]|2.22| No correlation found to any other genes in chemogenomics.
