Regulates the development, function and differentiation of conventional T- cells and nonconventional NKT-cells. When antigen presenting cells (APC) activate T-cell receptor (TCR), a series of phosphorylation lead to the recruitment of TXK to the cell membrane, where it is phosphorylated at Tyr-420. Phosphorylation leads to TXK full activation. Contributes also to signaling from many receptors and participates in multiple downstream pathways, including regulation of the actin cytoskeleton. Like ITK, can phosphorylate PLCG1, leading to its localization in lipid rafts and activation, followed by subsequent cleavage of its substrates. In turn, the endoplasmic reticulum releases calcium in the cytoplasm and the nuclear activator of activated T-cells (NFAT) translocates into the nucleus to perform its transcriptional duty. With PARP1 and EEF1A1, TXK forms a complex that acts as a T-helper 1 (Th1) cell- specific transcription factor and binds the promoter of IFNG to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production. Phosphorylates both PARP1 and EEF1A1. Phosphorylates also key sites in LCP2 leading to the up-regulation of Th1 preferred cytokine IL-2. Phosphorylates 'Tyr- 201' of CTLA4 which leads to the association of PI-3 kinase with the CTLA4 receptor. {ECO:0000269|PubMed:10523612, ECO:0000269|PubMed:11564877, ECO:0000269|PubMed:11859127, ECO:0000269|PubMed:17177976, ECO:0000269|PubMed:9813138}. |SH3 1| |Pkinase| |Pkinase Tyr| |SH2| |positive regulation of response to interferon-gamma| |positive regulation of interferon-gamma-mediated signaling pathway| |RNA polymerase II regulatory region DNA binding| |regulation of interferon-gamma-mediated signaling pathway| |regulation of response to interferon-gamma| |activation of phospholipase C activity| |peptidyl-tyrosine autophosphorylation| |positive regulation of phospholipase C activity| |regulation of phospholipase C activity| |non-membrane spanning protein tyrosine kinase activity| |positive regulation of cytokine-mediated signaling pathway| |positive regulation of response to cytokine stimulus| |positive regulation of phospholipase activity| |positive regulation of interferon-gamma production| |regulation of phospholipase activity| |positive regulation of lipase activity| |regulation of lipase activity| |regulation of interferon-gamma production| |peptidyl-tyrosine phosphorylation| |peptidyl-tyrosine modification| |cytokine production| |regulation of cytokine-mediated signaling pathway| |regulation of response to cytokine stimulus| |T cell receptor signaling pathway| |protein autophosphorylation| |antigen receptor-mediated signaling pathway| |positive regulation of innate immune response| |positive regulation of response to biotic stimulus| |DNA-binding transcription activator activity, RNA polymerase II-specific| |immune response-activating cell surface receptor signaling pathway| |positive regulation of cytokine production| |regulation of innate immune response| |positive regulation of defense response| |immune response-regulating cell surface receptor signaling pathway| |RNA polymerase II proximal promoter sequence-specific DNA binding| |positive regulation of multi-organism process| |regulation of response to biotic stimulus| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |positive regulation of response to external stimulus| |adaptive immune response| |activation of immune response| |regulation of cytokine production| |regulation of defense response| |positive regulation of hydrolase activity| |regulation of multi-organism process| |positive regulation of immune response| |peptidyl-amino acid modification| |protein phosphorylation| |regulation of response to external stimulus| |positive regulation of immune system process| |regulation of immune response| |positive regulation of transcription by RNA polymerase II| |regulation of hydrolase activity| |phosphorylation| |positive regulation of catalytic activity| |regulation of response to stress| |ATP binding| |positive regulation of transcription, DNA-templated| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |regulation of immune system process| |positive regulation of signal transduction| |positive regulation of RNA metabolic process| |positive regulation of multicellular organismal process| |positive regulation of molecular function| |positive regulation of cell communication| |positive regulation of signaling| |immune response| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === No hits were found.
