Catalyzes 'Lys-11'-linked polyubiquitination. Acts as an essential factor of the anaphase promoting complex/cyclosome (APC/C), a cell cycle- regulated ubiquitin ligase that controls progression through mitosis. Acts by specifically elongating 'Lys-11'-linked polyubiquitin chains initiated by the E2 enzyme UBE2C/UBCH10 on APC/C substrates, enhancing the degradation of APC/C substrates by the proteasome and promoting mitotic exit (PubMed:19820702, PubMed:19822757, PubMed:27259151). Also acts by elongating ubiquitin chains initiated by the E2 enzyme UBE2D1/UBCH5 in vitro; it is however unclear whether UBE2D1/UBCH5 acts as an E2 enzyme for the APC/C in vivo. Also involved in ubiquitination and subsequent degradation of VHL, resulting in an accumulation of HIF1A (PubMed:16819549). In vitro able to promote polyubiquitination using all 7 ubiquitin Lys residues, except 'Lys-48'-linked polyubiquitination (PubMed:20061386, PubMed:20622874). {ECO:0000269|PubMed:16819549, ECO:0000269|PubMed:19820702, ECO:0000269|PubMed:19822757, ECO:0000269|PubMed:20061386, ECO:0000269|PubMed:20622874, ECO:0000269|PubMed:22496338, ECO:0000269|PubMed:27259151}. |UQ con| |free ubiquitin chain polymerization| |protein K27-linked ubiquitination| |protein K29-linked ubiquitination| |ubiquitin recycling| |protein K6-linked ubiquitination| |anaphase-promoting complex binding| |positive regulation of ubiquitin protein ligase activity| |exit from mitosis| |anaphase-promoting complex| |regulation of ubiquitin protein ligase activity| |protein K11-linked ubiquitination| |positive regulation of ubiquitin-protein transferase activity| |protein K63-linked ubiquitination| |ubiquitin conjugating enzyme activity| |regulation of ubiquitin-protein transferase activity| |anaphase-promoting complex-dependent catabolic process| |protein polymerization| |positive regulation of protein ubiquitination| |positive regulation of protein modification by small protein conjugation or removal| |mitotic nuclear division| |regulation of protein ubiquitination| |regulation of protein modification by small protein conjugation or removal| |ubiquitin-protein transferase activity| |mitotic cell cycle phase transition| |cell cycle phase transition| |nuclear division| |protein polyubiquitination| |organelle fission| |proteasome-mediated ubiquitin-dependent protein catabolic process| |proteasomal protein catabolic process| |cell division| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |mitotic cell cycle process| |cellular protein catabolic process| |positive regulation of transferase activity| |mitotic cell cycle| |protein catabolic process| |protein ubiquitination| |protein modification by small protein conjugation| |cellular protein-containing complex assembly| |cellular homeostasis| |cellular macromolecule catabolic process| |regulation of transferase activity| |protein modification by small protein conjugation or removal| |cell cycle process| |macromolecule catabolic process| |organonitrogen compound catabolic process| |positive regulation of protein modification process| |proteolysis| |cell cycle| |positive regulation of catalytic activity| |ATP binding| |protein-containing complex assembly| |positive regulation of cellular protein metabolic process| |homeostatic process| |positive regulation of protein metabolic process| |organic substance catabolic process| |positive regulation of molecular function| |cellular catabolic process| |regulation of protein modification process| |protein-containing complex subunit organization| \\ === CRISPR Data === ^Screen^Score^ |Apcin 50 to 100μM on day4 R04 exp176|-4.33| |proTAME 5μM R04 exp187|-3.39| |Apcin 25μM plus proTAME 2μM R04 exp177|-3.24| |BI-2536 0.0042μM R03 exp95|-1.99| |Etoposide 0.1μM R05 exp198|-1.89| |TRAIL 50ng/ml R00 exp36|-1.79| |Centrinone 0.5μM R07 exp343|-1.78| |Latrunculin-B 10μM R07 exp374|-1.78| ^Gene^Correlation^ |UBE2C|0.566| |ANAPC13|0.5| |KIF20A|0.465| |MAD2L1BP|0.418| |SKA1|0.418| |ANAPC15|0.415| Global Fraction of Cell Lines Where Essential: 20/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|1/28| |blood|4/28| |bone|0/26| |breast|2/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|1/21| |liver|3/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|2/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|2/22| |urinary tract|0/29| |uterus|1/5| == Essentiality in NALM6 == * **Essentiality Rank**: 7886 * **Expression level (log2 read counts)**: 7.77
