Members of this family can be subdivided in two groups, the IRSp53-like and MIM-like, based on the presence or absence of the SH3 (Src homology 3) domain. The protein encoded by this gene contains a conserved IMD, also known as F-actin bundling domain, at the N-terminus, and a canonical SH3 domain near the C-terminus, so it belongs to the IRSp53-like group. This protein is the substrate for insulin receptor tyrosine kinase and binds to the small GTPase Rac. It is involved in signal transduction pathways that link deformation of the plasma membrane and remodeling of the actin cytoskeleton. It also promotes actin assembly and membrane protrusions when overexpressed in mammalian cells, and is essential to the formation of a potent actin assembly complex during EHEC (Enterohemorrhagic Escherichia coli) pedestal formation. [provided by RefSeq, Oct 2009]. * **<color #00a2e8>UniProt Summary</color>**: May function as adapter protein. Involved in the formation of clusters of actin bundles. Plays a role in the reorganization of the actin cytoskeleton in response to bacterial infection. {ECO:0000269|PubMed:17430976, ECO:0000269|PubMed:19366662, ECO:0000269|PubMed:22921828}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |IMD| |SH3 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |actin crosslink formation| |proline-rich region binding| |cadherin binding involved in cell-cell adhesion| |positive regulation of actin cytoskeleton reorganization| |regulation of actin cytoskeleton reorganization| |actin filament bundle assembly| |regulation of insulin receptor signaling pathway| |actin filament bundle organization| |regulation of cellular response to insulin stimulus| |positive regulation of actin filament polymerization| |plasma membrane organization| |cell-cell adherens junction| |positive regulation of protein polymerization| |regulation of actin filament polymerization| |regulation of actin polymerization or depolymerization| |regulation of actin filament length| |positive regulation of supramolecular fiber organization| |actin cytoskeleton| |positive regulation of cytoskeleton organization| |regulation of protein polymerization| |actin filament organization| |positive regulation of protein complex assembly| |regulation of actin filament organization| |actin binding| |regulation of actin cytoskeleton organization| |regulation of supramolecular fiber organization| |regulation of cellular component size| |regulation of actin filament-based process| |endomembrane system organization| |regulation of protein complex assembly| |supramolecular fiber organization| |actin cytoskeleton organization| |cell-cell adhesion| |regulation of anatomical structure size| |positive regulation of cellular component biogenesis| |regulation of cytoskeleton organization| |actin filament-based process| |positive regulation of organelle organization| |response to bacterium| |membrane organization| |cell adhesion| |biological adhesion| |regulation of cellular component biogenesis| |cytoskeleton organization| |positive regulation of cellular component organization| |regulation of organelle organization| |response to other organism| |response to external biotic stimulus| |response to biotic stimulus| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Mubritinib 0.2μM R02 exp65|-1.86| |CFI-400945 25μM R08 exp469|-1.73| |Vitamin-D3 40μM R08 exp536|-1.72| |Metformin 100 to 150μM on day4 R04 exp170|1.72| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 12540 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 0.43 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='BAIAP2L1 Expression in NALM6 Cells: 0.43'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
