The protein has a role in B-cell receptor signaling and B-cell development. The protein also stimulates insulin synthesis and secretion in response to glucose and enhances the expression of several pancreatic beta-cell transcription factors. [provided by RefSeq, Aug 2010]. * **<color #00a2e8>UniProt Summary</color>**: Non-receptor tyrosine kinase involved in B-lymphocyte development, differentiation and signaling. B-cell receptor (BCR) signaling requires a tight regulation of several protein tyrosine kinases and phosphatases, and associated coreceptors. Binding of antigen to the B-cell antigen receptor (BCR) triggers signaling that ultimately leads to B-cell activation. Signaling through BLK plays an important role in transmitting signals through surface immunoglobulins and supports the pro-B to pre-B transition, as well as the signaling for growth arrest and apoptosis downstream of B-cell receptor. Specifically binds and phosphorylates CD79A at 'Tyr-188'and 'Tyr-199', as well as CD79B at 'Tyr-196' and 'Tyr- 207'. Phosphorylates also the immunoglobulin G receptors FCGR2A, FCGR2B and FCGR2C. With FYN and LYN, plays an essential role in pre-B-cell receptor (pre-BCR)-mediated NF-kappa-B activation. Contributes also to BTK activation by indirectly stimulating BTK intramolecular autophosphorylation. In pancreatic islets, acts as a modulator of beta-cells function through the up-regulation of PDX1 and NKX6-1 and consequent stimulation of insulin secretion in response to glucose. {ECO:0000269|PubMed:19667185, ECO:0000269|PubMed:8756631}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Pkinase Tyr| |SH2| |Pkinase| |SH3 1| |SH3 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |regulation of B cell receptor signaling pathway| |peptidyl-tyrosine autophosphorylation| |non-membrane spanning protein tyrosine kinase activity| |extrinsic component of cytoplasmic side of plasma membrane| |regulation of antigen receptor-mediated signaling pathway| |positive regulation of insulin secretion| |protein tyrosine kinase activity| |positive regulation of peptide hormone secretion| |B cell receptor signaling pathway| |positive regulation of hormone secretion| |peptidyl-tyrosine phosphorylation| |peptidyl-tyrosine modification| |regulation of insulin secretion| |protein autophosphorylation| |regulation of peptide hormone secretion| |regulation of hormone secretion| |positive regulation of protein secretion| |positive regulation of peptide secretion| |antigen receptor-mediated signaling pathway| |signaling receptor binding| |positive regulation of secretion by cell| |positive regulation of protein transport| |positive regulation of secretion| |immune response-activating cell surface receptor signaling pathway| |positive regulation of establishment of protein localization| |regulation of protein secretion| |immune response-regulating cell surface receptor signaling pathway| |regulation of peptide secretion| |transmembrane receptor protein tyrosine kinase signaling pathway| |regulation of hormone levels| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |activation of immune response| |regulation of protein transport| |enzyme linked receptor protein signaling pathway| |regulation of peptide transport| |regulation of establishment of protein localization| |regulation of secretion by cell| |regulation of secretion| |positive regulation of immune response| |peptidyl-amino acid modification| |protein phosphorylation| |positive regulation of transport| |regulation of protein localization| |positive regulation of immune system process| |regulation of immune response| |phosphorylation| |ATP binding| |regulation of immune system process| |intracellular signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |regulation of transport| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 4129 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.24 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='BLK Expression in NALM6 Cells: 6.24'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
