This protein is stored in the zymogen granules of the nuclear envelope and functions by cleaving DNA in an endonucleolytic manner. At least six autosomal codominant alleles have been characterized, DNASE1*1 through DNASE1*6, and the sequence of DNASE1*2 represented in this record. Mutations in this gene have been associated with systemic lupus erythematosus (SLE), an autoimmune disease. A recombinant form of this protein is used to treat the one of the symptoms of cystic fibrosis by hydrolyzing the extracellular DNA in sputum and reducing its viscosity. Alternate transcriptional splice variants of this gene have been observed but have not been thoroughly characterized. Expressed by non-hematopoietic tissues and preferentially cleaves protein-free DNA (By similarity). Among other functions, seems to be involved in cell death by apoptosis (PubMed:11241278). Binds specifically to G- actin and blocks actin polymerization (By similarity). Together with DNASE1L3, plays a key role in degrading neutrophil extracellular traps (NETs) (By similarity). NETs are mainly composed of DNA fibers and are released by neutrophils to bind pathogens during inflammation (By similarity). Degradation of intravascular NETs by DNASE1 and DNASE1L3 is required to prevent formation of clots that obstruct blood vessels and cause organ damage following inflammation (By similarity). {ECO:0000250|UniProtKB:P00639, ECO:0000250|UniProtKB:P21704, ECO:0000250|UniProtKB:P49183, ECO:0000269|PubMed:11241278, ECO:0000269|PubMed:2251263, ECO:0000269|PubMed:2277032}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Exo endo phos| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |regulation of neutrophil mediated cytotoxicity| |deoxyribonuclease I activity| |deoxyribonuclease activity| |DNA catabolic process, endonucleolytic| |DNA catabolic process| |regulation of acute inflammatory response| |regulation of myeloid leukocyte mediated immunity| |regulation of leukocyte mediated cytotoxicity| |regulation of cell killing| |nuclear envelope| |regulation of leukocyte mediated immunity| |actin binding| |nucleic acid phosphodiester bond hydrolysis| |regulation of inflammatory response| |nucleobase-containing compound catabolic process| |heterocycle catabolic process| |cellular nitrogen compound catabolic process| |aromatic compound catabolic process| |regulation of immune effector process| |organic cyclic compound catabolic process| |neutrophil activation involved in immune response| |neutrophil activation| |granulocyte activation| |myeloid cell activation involved in immune response| |myeloid leukocyte activation| |leukocyte activation involved in immune response| |cell activation involved in immune response| |DNA metabolic process| |regulation of defense response| |cellular macromolecule catabolic process| |apoptotic process| |leukocyte activation| |macromolecule catabolic process| |programmed cell death| |cell activation| |immune effector process| |cell death| |regulation of response to external stimulus| |DNA binding| |regulation of response to stress| |regulation of immune system process| |organic substance catabolic process| |cellular catabolic process| |immune response| |extracellular region| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp379|MSC2530818 10μM R07 exp379]]|1.74| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |[[:human genes:r:rrm1|RRM1]]|0.626| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 2805 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.87 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='DNASE1 Expression in NALM6 Cells: 5.87'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
