The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2:GTP:methionyl-tRNAi and eIF-5 to form the 43S pre- initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation (PubMed:17581632). The eIF- 3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed:25849773). {ECO:0000255|HAMAP-Rule:MF_03011, ECO:0000269|PubMed:17581632, ECO:0000269|PubMed:25849773, ECO:0000269|PubMed:27462815}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Paf67| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |viral translational termination-reinitiation| |viral translation| |eukaryotic 48S preinitiation complex| |formation of cytoplasmic translation initiation complex| |eukaryotic translation initiation factor 3 complex| |eukaryotic 43S preinitiation complex| |cytoplasmic translational initiation| |translation initiation factor activity| |cytoplasmic translation| |viral gene expression| |translational initiation| |ribonucleoprotein complex assembly| |ribonucleoprotein complex subunit organization| |translation| |peptide biosynthetic process| |ribonucleoprotein complex biogenesis| |amide biosynthetic process| |peptide metabolic process| |viral process| |symbiotic process| |cellular amide metabolic process| |interspecies interaction between organisms| |cellular protein-containing complex assembly| |RNA binding| |organonitrogen compound biosynthetic process| |protein-containing complex assembly| |cellular nitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |protein-containing complex subunit organization| |membrane| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |PU-H71 1μM R08 exp515|-2.65| |MS023 2μM R03 exp134|-2.59| |BVD-523 15μM R02 exp67|-2.22| |SNS-032 15μM R08 exp443|-2.18| |Staurosporine 0.02μM R08 exp524|-2.04| |Pyridostatin 4μM R05 exp240|-1.94| |Tozasertib 0.1μM R07 exp414|-1.85| |GSK626616 14μM R08 exp485|-1.73| |Rucaparib 6.5μM R08 exp520|-1.73| |Salinomycin 3.5μM R08 exp521|-1.73| |Dorsomorphin 5μM R07 exp357|-1.7| |Fluvastatin 2.2μM R06 exp282|1.74| |Epothilone-B 0.0015μM R05 exp231|1.82| |I-BRD9 4μM R07 exp365|2.03| |TRAIL 50ng/ml R00 exp36|2.13| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |EIF3K|0.657| |ATP13A1|0.488| |BCL2L1|0.467| |SMG7|0.412| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 7/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|1/56| |cervix|0/4| |colorectal|0/17| |esophagus|1/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|1/16| |ovary|0/26| |pancreas|1/24| |peripheral nervous system|0/16| |plasma cell|2/15| |prostate|0/1| |skin|1/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 2154 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 8.13 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='EIF3L Expression in NALM6 Cells: 8.13'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
