NMDA channel has been shown to be involved in long-term potentiation, an activity-dependent increase in the efficiency of synaptic transmission thought to underlie certain kinds of memory and learning. NMDA receptor channels are heteromers composed of the key receptor subunit NMDAR1 (GRIN1) and 1 or more of the 4 NMDAR2 subunits: NMDAR2A (GRIN2A), NMDAR2B (GRIN2B), NMDAR2C (GRIN2C), and NMDAR2D (GRIN2D). [provided by RefSeq, Mar 2010]. * **<color #00a2e8>UniProt Summary</color>**: Component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Channel activation requires binding of the neurotransmitter glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+) (PubMed:9489750, PubMed:27616483, PubMed:26875626, PubMed:28126851). Sensitivity to glutamate and channel kinetics depend on the subunit composition (PubMed:9489750). {ECO:0000269|PubMed:26875626, ECO:0000269|PubMed:27616483, ECO:0000269|PubMed:28095420, ECO:0000269|PubMed:28126851, ECO:0000269|PubMed:9489750}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |ANF receptor| |Lig chan-Glu bd| |SBP bac 3| |Lig chan| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |glutamate-gated calcium ion channel activity| |NMDA glutamate receptor activity| |ionotropic glutamate receptor activity| |excitatory chemical synaptic transmission| |NMDA selective glutamate receptor complex| |postsynaptic density membrane| |ionotropic glutamate receptor signaling pathway| |startle response| |regulation of sensory perception of pain| |regulation of sensory perception| |glutamate receptor signaling pathway| |long-term synaptic potentiation| |excitatory postsynaptic potential| |calcium ion transmembrane import into cytosol| |chemical synaptic transmission, postsynaptic| |calcium ion transport into cytosol| |adult locomotory behavior| |cytosolic calcium ion transport| |regulation of postsynaptic membrane potential| |neuromuscular process| |regulation of nervous system process| |adult behavior| |calcium-mediated signaling| |positive regulation of synaptic transmission| |regulation of synaptic plasticity| |locomotory behavior| |calcium ion transmembrane transport| |calcium ion transport| |positive regulation of cytosolic calcium ion concentration| |divalent metal ion transport| |divalent inorganic cation transport| |regulation of cytosolic calcium ion concentration| |second-messenger-mediated signaling| |chemical synaptic transmission| |anterograde trans-synaptic signaling| |cell surface receptor signaling pathway involved in cell-cell signaling| |regulation of membrane potential| |cellular calcium ion homeostasis| |trans-synaptic signaling| |modulation of chemical synaptic transmission| |regulation of trans-synaptic signaling| |calcium ion homeostasis| |synaptic signaling| |cellular divalent inorganic cation homeostasis| |divalent inorganic cation homeostasis| |cell junction| |cellular metal ion homeostasis| |inorganic cation transmembrane transport| |behavior| |regulation of system process| |cation transmembrane transport| |metal ion homeostasis| |cellular cation homeostasis| |metal ion transport| |cellular ion homeostasis| |inorganic ion transmembrane transport| |cation homeostasis| |inorganic ion homeostasis| |brain development| |cellular chemical homeostasis| |head development| |ion homeostasis| |cation transport| |cellular homeostasis| |ion transmembrane transport| |central nervous system development| |chemical homeostasis| |cell-cell signaling| |transmembrane transport| |ion transport| |nervous system process| |integral component of plasma membrane| |homeostatic process| |intracellular signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |establishment of localization in cell| |system process| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 8285 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -1.04 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='GRIN2D Expression in NALM6 Cells: -1.04'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
