The encoded protein binds numerous other proteins including group I metabotropic glutamate receptors, inositol 1,4,5-trisphosphate receptors and amyloid precursor proteins and has been implicated in diverse biological functions such as neuronal signaling, T-cell activation and trafficking of amyloid beta peptides. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Mar 2009]. * **<color #00a2e8>UniProt Summary</color>**: Postsynaptic density scaffolding protein. Binds and cross-links cytoplasmic regions of GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1 and SHANK3. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, it aids the coupling of surface receptors to intracellular calcium release. Isoforms can be differently regulated and may play an important role in maintaining the plasticity at glutamatergic synapses. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |WH1| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |G protein-coupled glutamate receptor binding| |regulation of store-operated calcium entry| |negative regulation of calcineurin-mediated signaling| |negative regulation of calcineurin-NFAT signaling cascade| |G protein-coupled glutamate receptor signaling pathway| |negative regulation of interleukin-2 production| |negative regulation of calcium-mediated signaling| |regulation of calcineurin-NFAT signaling cascade| |regulation of calcineurin-mediated signaling| |glutamate receptor signaling pathway| |regulation of interleukin-2 production| |regulation of calcium-mediated signaling| |postsynaptic membrane| |regulation of calcium ion transport| |postsynaptic density| |negative regulation of cytokine production| |neuron projection| |protein targeting| |regulation of metal ion transport| |dendrite| |cellular component| |negative regulation of intracellular signal transduction| |cell junction| |regulation of cytokine production| |regulation of ion transport| |intracellular protein transport| |identical protein binding| |negative regulation of multicellular organismal process| |negative regulation of signal transduction| |G protein-coupled receptor signaling pathway| |negative regulation of cell communication| |negative regulation of signaling| |protein transport| |intracellular transport| |peptide transport| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |negative regulation of response to stimulus| |regulation of intracellular signal transduction| |establishment of localization in cell| |nitrogen compound transport| |regulation of transport| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp365|I-BRD9 4μM R07 exp365]]|1.74| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5244 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 1.25 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='HOMER3 Expression in NALM6 Cells: 1.25'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
