The kelch motif appears in many different polypeptide contexts and contains multiple potential protein-protein contact sites. Members of this family are present both throughout the cell and extracellularly, with diverse activities. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Substrate-specific adapter of a BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex involved in interferon response and anterograde Golgi to endosome transport. The BCR(KLHL20) E3 ubiquitin ligase complex mediates the ubiquitination of DAPK1, leading to its degradation by the proteasome, thereby acting as a negative regulator of apoptosis (PubMed:20389280). The BCR(KLHL20) E3 ubiquitin ligase complex also specifically mediates 'Lys-33'- linked ubiquitination (PubMed:24768539). Involved in anterograde Golgi to endosome transport by mediating 'Lys-33'-linked ubiquitination of CORO7, promoting interaction between CORO7 and EPS15, thereby facilitating actin polymerization and post-Golgi trafficking (PubMed:24768539). Also acts as a regulator of endothelial migration during angiogenesis by controlling the activation of Rho GTPases. The BCR(KLHL20) E3 ubiquitin ligase complex acts as a regulator of neurite outgrowth by mediating ubiquitination and degradation of PDZ-RhoGEF/ARHGEF11 (PubMed:21670212). In case of tumor, the BCR(KLHL20) E3 ubiquitin ligase complex is involved in tumor hypoxia: following hypoxia, the BCR(KLHL20)complex mediates ubiquitination and degradation of PML, potentiating HIF-1 signaling and cancer progression (PubMed:21840486). {ECO:0000269|PubMed:14528312, ECO:0000269|PubMed:17395875, ECO:0000269|PubMed:20389280, ECO:0000269|PubMed:21670212, ECO:0000269|PubMed:21840486, ECO:0000269|PubMed:24768539}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |BACK| |Kelch 1| |BTB| |Kelch 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |protein K33-linked ubiquitination| |interferon-gamma binding| |Golgi to endosome transport| |response to interferon-alpha| |Cul3-RING ubiquitin ligase complex| |PML body| |post-Golgi vesicle-mediated transport| |cytosolic transport| |trans-Golgi network| |actin cytoskeleton| |ubiquitin-protein transferase activity| |actin binding| |axon| |protein polyubiquitination| |proteasome-mediated ubiquitin-dependent protein catabolic process| |proteasomal protein catabolic process| |post-translational protein modification| |Golgi vesicle transport| |dendrite| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |protein catabolic process| |protein ubiquitination| |perinuclear region of cytoplasm| |protein modification by small protein conjugation| |negative regulation of apoptotic process| |negative regulation of programmed cell death| |cellular macromolecule catabolic process| |Golgi apparatus| |protein modification by small protein conjugation or removal| |negative regulation of cell death| |macromolecule catabolic process| |organonitrogen compound catabolic process| |response to cytokine| |cytoskeleton organization| |proteolysis| |protein transport| |intracellular transport| |peptide transport| |regulation of apoptotic process| |regulation of programmed cell death| |amide transport| |establishment of protein localization| |regulation of cell death| |organic substance catabolic process| |cellular catabolic process| |establishment of localization in cell| |nitrogen compound transport| |vesicle-mediated transport| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |GSK626616 14μM R08 exp485|-1.77| |Phorbol-12-myristate-13-acetate 0.57μM R08 exp514|-1.77| |Cerivastatin 0.15μM R05 exp226|-1.74| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 11573 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 4.98 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='KLHL20 Expression in NALM6 Cells: 4.98'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
