This protein also associates with N-oligosaccharyl transferase and therefore may have a role in N-glycosylation. Mutations in this gene cause a form of X-linked intellectual disability (XLID). This gene may have multiple in-frame translation initiation sites, one of which would encode a shorter protein with an N-terminus containing a signal peptide at amino acids 1-29. [provided by RefSeq, Jul 2017]. * **<color #00a2e8>UniProt Summary</color>**: Acts as accessory component of the N-oligosaccharyl transferase (OST) complex which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains. Involved in N-glycosylation of STT3B-dependent substrates. Specifically required for the glycosylation of a subset of acceptor sites that are near cysteine residues; in this function seems to act redundantly with TUSC3. In its oxidized form proposed to form transient mixed disulfides with a glycoprotein substrate to facilitate access of STT3B to the unmodified acceptor site. Has also oxidoreductase-independent functions in the STT3B-containing OST complex possibly involving substrate recognition. {ECO:0000269|PubMed:25135935, ECO:0000269|PubMed:26864433, ECO:0000305}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |OST3 OST6| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |dolichyl-diphosphooligosaccharide-protein glycotransferase activity| |oligosaccharyltransferase complex| |magnesium ion transmembrane transporter activity| |magnesium ion transmembrane transport| |magnesium ion transport| |protein N-linked glycosylation via asparagine| |peptidyl-asparagine modification| |azurophil granule membrane| |protein N-linked glycosylation| |protein glycosylation| |macromolecule glycosylation| |glycosylation| |divalent metal ion transport| |cognition| |divalent inorganic cation transport| |glycoprotein biosynthetic process| |glycoprotein metabolic process| |neutrophil degranulation| |neutrophil activation involved in immune response| |neutrophil mediated immunity| |neutrophil activation| |granulocyte activation| |leukocyte degranulation| |myeloid leukocyte mediated immunity| |myeloid cell activation involved in immune response| |inorganic cation transmembrane transport| |myeloid leukocyte activation| |cation transmembrane transport| |carbohydrate derivative biosynthetic process| |leukocyte activation involved in immune response| |cell activation involved in immune response| |metal ion transport| |inorganic ion transmembrane transport| |regulated exocytosis| |leukocyte mediated immunity| |exocytosis| |cation transport| |peptidyl-amino acid modification| |leukocyte activation| |ion transmembrane transport| |secretion by cell| |endoplasmic reticulum| |carbohydrate derivative metabolic process| |export from cell| |cell activation| |immune effector process| |secretion| |transmembrane transport| |ion transport| |nervous system process| |integral component of plasma membrane| |organonitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |immune response| |vesicle-mediated transport| |system process| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Bortezomib 0.005μM R00 exp6|-2.18| |Mitomycin-C 0.06μM R08 exp503|-1.8| |Sulforaphane 9μM R08 exp526 no dilution day6|1.75| |Rapamycin 10μM R00 exp30|1.87| |Hippuristanol 0.12μM R08 exp488|1.89| |Bafilomycin-A1 0.009μM R08 exp454|2.5| |Lead acetate 2000μM R08 exp497|2.5| |Lead acetate 2000μM R08 exp498 no dilution day6|2.71| |2-Deoxy-D-glucose 800μM R05 exp214|2.91| |Arsenate 40μM R06 exp264|3.79| |Thapsigargin 0.005μM R07 exp407|4.29| |Tunicamycin 0.04μM R07 exp418|4.87| |Tunicamycin 0.04 to 0.125μM on day4 R07 exp420|5.96| |Tunicamycin 0.04 to 0.075μM on day4 R07 exp419|6.58| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |STT3B|0.651| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/694 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/26| |bone|0/26| |breast|0/30| |central nervous system|0/49| |cervix|0/4| |colorectal|0/17| |esophagus|0/11| |fibroblast|0/1| |gastric|0/14| |kidney|0/18| |liver|0/19| |lung|0/72| |lymphocyte|0/16| |ovary|0/25| |pancreas|0/22| |peripheral nervous system|0/15| |plasma cell|0/12| |prostate|0/1| |skin|0/20| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/28| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 12739 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.89 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='MAGT1 Expression in NALM6 Cells: 6.89'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
