The mediator complex is recruited by transcriptional activators or nuclear receptors to induce gene expression, by interacting with RNA polymerase II and promoting the formation of a transcriptional pre-initiation complex. A mutation in this gene has been associated with a novel infantile-onset neurodegenerative movement disorder. Alternatively spliced transcript variants have been identified. [provided by RefSeq, Mar 2015]. * **<color #00a2e8>UniProt Summary</color>**: Component of the Mediator complex, a coactivator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Med20| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |DNA-directed 5-3 RNA polymerase activity| |mediator complex| |ubiquitin ligase complex| |transcription initiation from RNA polymerase II promoter| |DNA-templated transcription, initiation| |ubiquitin protein ligase activity| |transcription coactivator activity| |transcription by RNA polymerase II| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |protein ubiquitination| |protein modification by small protein conjugation| |protein modification by small protein conjugation or removal| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |organic cyclic compound biosynthetic process| |cellular nitrogen compound biosynthetic process| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |RNA metabolic process| |cellular macromolecule biosynthetic process| |positive regulation of RNA metabolic process| |macromolecule biosynthetic process| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |gene expression| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp380|NMS-873 0.07μM R07 exp380]]|-2| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 22/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|1/28| |blood|1/28| |bone|2/25| |breast|2/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/15| |kidney|1/21| |liver|0/20| |lung|2/75| |lymphocyte|0/14| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|1/16| |plasma cell|1/15| |prostate|0/1| |skin|1/24| |soft tissue|0/7| |thyroid|0/2| |upper aerodigestive|1/22| |urinary tract|1/29| |uterus|1/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1495 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.05 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='MED20 Expression in NALM6 Cells: 5.05'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
