It is an integral component of the pericentriolar material (PCM). The protein contains a series of coiled-coil domains and a highly conserved PCM targeting motif called the PACT domain near its C-terminus. The protein interacts with the microtubule nucleation component gamma-tubulin and is likely important to normal functioning of the centrosomes, cytoskeleton, and cell-cycle progression. Mutations in this gene cause Seckel syndrome-4 and microcephalic osteodysplastic primordial dwarfism type II. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2015]. * **<color #00a2e8>UniProt Summary</color>**: Integral component of the filamentous matrix of the centrosome involved in the initial establishment of organized microtubule arrays in both mitosis and meiosis. Plays a role, together with DISC1, in the microtubule network formation. Is an integral component of the pericentriolar material (PCM). May play an important role in preventing premature centrosome splitting during interphase by inhibiting NEK2 kinase activity at the centrosome. {ECO:0000269|PubMed:10823944, ECO:0000269|PubMed:11171385, ECO:0000269|PubMed:18955030, ECO:0000269|PubMed:20599736}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |PACT coil coil| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |centriolar satellite| |molecular adaptor activity| |mitotic spindle organization| |ciliary basal body-plasma membrane docking| |microtubule cytoskeleton organization involved in mitosis| |G2/M transition of mitotic cell cycle| |cell cycle G2/M phase transition| |centriole| |positive regulation of intracellular protein transport| |spindle organization| |microtubule organizing center| |organelle localization by membrane tethering| |membrane docking| |regulation of G2/M transition of mitotic cell cycle| |calmodulin binding| |positive regulation of intracellular transport| |regulation of cell cycle G2/M phase transition| |regulation of intracellular protein transport| |mitotic cell cycle phase transition| |cell cycle phase transition| |microtubule| |positive regulation of cellular protein localization| |cilium assembly| |regulation of intracellular transport| |cilium organization| |regulation of mitotic cell cycle phase transition| |positive regulation of protein transport| |plasma membrane bounded cell projection assembly| |regulation of cell cycle phase transition| |cell projection assembly| |positive regulation of establishment of protein localization| |microtubule cytoskeleton organization| |centrosome| |regulation of cellular protein localization| |organelle localization| |mitotic cell cycle process| |regulation of mitotic cell cycle| |microtubule-based process| |mitotic cell cycle| |regulation of protein transport| |regulation of peptide transport| |regulation of establishment of protein localization| |regulation of cell cycle process| |organelle assembly| |regulation of cellular localization| |positive regulation of transport| |cell cycle process| |regulation of protein localization| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |cell projection organization| |regulation of cell cycle| |cell cycle| |regulation of transport| |membrane| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 2/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|1/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1956 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.95 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PCNT Expression in NALM6 Cells: 7.95'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
