L-serine is required for D-serine and other amino acid synthesis. The enzyme requires NAD/NADH as a cofactor and forms homotetramers for activity. Mutations in this gene have been found in a family with congenital microcephaly, psychomotor retardation and other symptoms. Multiple alternatively spliced transcript variants have been found, however the full-length nature of most are not known. [provided by RefSeq, Aug 2011]. * **<color #00a2e8>UniProt Summary</color>**: Catalyzes the reversible oxidation of 3-phospho-D- glycerate to 3-phosphonooxypyruvate, the first step of the phosphorylated L-serine biosynthesis pathway. Also catalyzes the reversible oxidation of 2-hydroxyglutarate to 2-oxoglutarate and the reversible oxidation of (S)-malate to oxaloacetate. {ECO:0000269|PubMed:11751922, ECO:0000269|PubMed:25406093}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |2-Hacid dh C| |2-Hacid dh| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |phosphoglycerate dehydrogenase activity| |L-malate dehydrogenase activity| |gamma-aminobutyric acid metabolic process| |threonine metabolic process| |L-serine biosynthetic process| |alkanesulfonate metabolic process| |taurine metabolic process| |G1 to G0 transition| |L-serine metabolic process| |serine family amino acid biosynthetic process| |glycine metabolic process| |glutamine metabolic process| |NAD binding| |serine family amino acid metabolic process| |aspartate family amino acid metabolic process| |alpha-amino acid biosynthetic process| |glutamine family amino acid metabolic process| |electron transfer activity| |cellular amino acid biosynthetic process| |neurotransmitter metabolic process| |glial cell development| |spinal cord development| |xenobiotic metabolic process| |neural tube development| |glial cell differentiation| |electron transport chain| |cellular response to xenobiotic stimulus| |alpha-amino acid metabolic process| |gliogenesis| |response to xenobiotic stimulus| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |cellular amino acid metabolic process| |regulation of neurotransmitter levels| |sulfur compound metabolic process| |generation of precursor metabolites and energy| |drug metabolic process| |monocarboxylic acid metabolic process| |small molecule biosynthetic process| |chordate embryonic development| |embryo development ending in birth or egg hatching| |neuron projection development| |brain development| |head development| |neuron development| |tube development| |carboxylic acid metabolic process| |oxidation-reduction process| |embryo development| |central nervous system development| |cell cycle process| |oxoacid metabolic process| |neuron differentiation| |organic acid metabolic process| |epithelium development| |plasma membrane bounded cell projection organization| |cell projection organization| |cell cycle| |organonitrogen compound biosynthetic process| |generation of neurons| |neurogenesis| |cell development| |small molecule metabolic process| |tissue development| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Mubritinib 0.2μM R01 exp48|-1.88| |A-395 10μM R05 exp218|-1.71| |Artemisinin 50μM R08 exp450|1.75| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 1/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/25| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/15| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/14| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/7| |thyroid|0/2| |upper aerodigestive|1/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 9212 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.49 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PHGDH Expression in NALM6 Cells: 7.49'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
