Protein phosphatase 2A is one of the four major Ser/Thr phosphatases, and it is implicated in the negative control of cell growth and division. It consists of a common heteromeric core enzyme, which is composed of a catalytic subunit and a constant regulatory subunit, that associates with a variety of regulatory subunits. The B regulatory subunit might modulate substrate selectivity and catalytic activity. This gene encodes a gamma isoform of the regulatory subunit B56 subfamily. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: The B regulatory subunit might modulate substrate selectivity and catalytic activity, and also might direct the localization of the catalytic enzyme to a particular subcellular compartment. The PP2A-PPP2R5C holoenzyme may specifically dephosphorylate and activate TP53 and play a role in DNA damage- induced inhibition of cell proliferation. PP2A-PPP2R5C may also regulate the ERK signaling pathway through ERK dephosphorylation. {ECO:0000269|PubMed:16456541, ECO:0000269|PubMed:17245430}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |B56| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |protein phosphatase activator activity| |protein phosphatase type 2A complex| |protein phosphatase regulator activity| |negative regulation of protein kinase B signaling| |regulation of protein autophosphorylation| |chromosome, centromeric region| |regulation of phosphoprotein phosphatase activity| |regulation of phosphatidylinositol 3-kinase signaling| |regulation of protein dephosphorylation| |regulation of phosphatase activity| |regulation of dephosphorylation| |protein dephosphorylation| |regulation of protein kinase B signaling| |dephosphorylation| |proteasome-mediated ubiquitin-dependent protein catabolic process| |proteasomal protein catabolic process| |negative regulation of intracellular signal transduction| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |negative regulation of cell population proliferation| |protein catabolic process| |cellular macromolecule catabolic process| |Golgi apparatus| |macromolecule catabolic process| |organonitrogen compound catabolic process| |negative regulation of signal transduction| |proteolysis| |regulation of hydrolase activity| |negative regulation of cell communication| |negative regulation of signaling| |regulation of protein phosphorylation| |regulation of phosphorylation| |regulation of cell population proliferation| |negative regulation of response to stimulus| |organic substance catabolic process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |cellular catabolic process| |regulation of intracellular signal transduction| |regulation of protein modification process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Taxol 0.004μM R03 exp103|-3.15| |BI-6727 0.0125μM R03 exp97|-2.79| |BI-2536 0.0042μM R03 exp95|-2.62| |Ixabepilone 0.004 to 0.005μM on day4 R04 exp184|-1.84| |GSK461364A 0.005μM R03 exp125|-1.75| |Phorbol-12-myristate-13-acetate 0.57μM R08 exp514|-1.72| |UMK57 0.6μM R03 exp107|1.78| |Staurosporine 0.02μM R08 exp524|2.05| |UMK57 1μM R01 exp59|2.48| |Mdivi-1 15μM R05 exp217|2.8| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |PPP2R1A|0.516| |LCMT1|0.49| |KIF18B|0.481| |KIF2C|0.463| |PPME1|0.427| |SNRK|0.423| |PPP2R5D|0.4| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 12245 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.61 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PPP2R5C Expression in NALM6 Cells: 7.61'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
