Structural domains of this protein include the catalytic diffuse B-cell lymphoma homology and pleckstrin homology (DHPH) domain, two disheveled, EGL-10, and pleckstrin homology (DEP) domains, two PDZ domains, and a C-terminal inositol polyphosphate-4 phosphatase (IP4P) domain that is found in one of the isoforms. This protein facilitates the exchange of GDP for GTP on Rac1, allowing the GTP-bound Rac1 to activate downstream effectors. Studies also show that the pleckstrin homology domain of this protein interacts with the phosphatase and tensin homolog (PTEN) gene product to inhibit PTEN phosphatase activity, thus activating the phosphoinositide-3 kinase (PI3K) signaling pathway. Conversely, the PTEN gene product has also been shown to inhibit the GEF activity of this protein. This gene plays a role in insulin-signaling pathways, and either mutations or overexpression of this gene have been observed in some cancers. [provided by RefSeq, Apr 2016]. * **<color #00a2e8>UniProt Summary</color>**: Functions as a RAC1 guanine nucleotide exchange factor (GEF), activating Rac proteins by exchanging bound GDP for free GTP. Its activity is synergistically activated by phosphatidylinositol 3,4,5-trisphosphate and the beta gamma subunits of heterotrimeric G protein. Mediates the activation of RAC1 in a PI3K-dependent manner. May be an important mediator of Rac signaling, acting directly downstream of both G protein- coupled receptors and phosphoinositide 3-kinase. {ECO:0000269|PubMed:15304342, ECO:0000269|PubMed:15304343, ECO:0000269|PubMed:15897194}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |DEP| |RhoGEF| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |Rac guanyl-nucleotide exchange factor activity| |phosphatidylinositol 3-kinase signaling| |dendrite morphogenesis| |phosphatidylinositol-mediated signaling| |inositol lipid-mediated signaling| |adult locomotory behavior| |dendrite development| |regulation of Rho protein signal transduction| |adult behavior| |locomotory behavior| |regulation of Ras protein signal transduction| |GTPase activator activity| |regulation of small GTPase mediated signal transduction| |positive regulation of GTPase activity| |cell morphogenesis involved in neuron differentiation| |neuron projection morphogenesis| |regulation of GTPase activity| |plasma membrane bounded cell projection morphogenesis| |cell projection morphogenesis| |cell part morphogenesis| |cell morphogenesis involved in differentiation| |behavior| |neuron projection development| |cell morphogenesis| |positive regulation of hydrolase activity| |neuron development| |cellular component morphogenesis| |neuron differentiation| |plasma membrane bounded cell projection organization| |cell projection organization| |regulation of hydrolase activity| |G protein-coupled receptor signaling pathway| |positive regulation of catalytic activity| |generation of neurons| |neurogenesis| |cell development| |intracellular signal transduction| |positive regulation of molecular function| |regulation of intracellular signal transduction| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Brefeldin A 0.02μM R00 exp8|1.71| |Nicotinate 1000μM R03 exp140|2.18| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |POLR2J3|0.514| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 8726 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 1.58 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PREX2 Expression in NALM6 Cells: 1.58'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
