Zinc finger domains of the encoded protein are required for its interaction with RIP and for inhibition of TNF- and IL1-induced NF-kappa B activation pathways. The encoded protein may also function as an E3 ubiquitin-protein ligase which accepts ubiquitin from E2 ubiquitin-conjugating enzymes and transfers it to substrates. Several alternatively spliced transcript variants have been described for this locus but the full-length natures of only some are known. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Isoform 1 acts as an E3 ubiquitin ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their degradation by the proteasome. Promotes degradation of TRAF3, TLR4 and TLR9. Contributes to the regulation of antiviral responses. Down- regulates activation of NF-kappa-B, IRF3 activation and IFNB production. Isoform 3 inhibits TNF and IL-1 mediated activation of NF-kappa-B. Promotes TNF and RIP mediated apoptosis. {ECO:0000269|PubMed:15107846, ECO:0000269|PubMed:19893624}. <button type='default' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> No Pfam Domain information is available for this gene. </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |protein catabolic process at synapse| |protein catabolic process, modulating synaptic transmission| |postsynaptic endocytic zone| |regulation of defense response to virus by host| |negative regulation of type I interferon production| |regulation of interferon-beta production| |protein K48-linked ubiquitination| |regulation of defense response to virus| |Schaffer collateral - CA1 synapse| |regulation of type I interferon production| |ubiquitin protein ligase activity| |negative regulation of cytokine production| |protein polyubiquitination| |proteasome-mediated ubiquitin-dependent protein catabolic process| |glutamatergic synapse| |proteasomal protein catabolic process| |modulation of chemical synaptic transmission| |regulation of trans-synaptic signaling| |regulation of immune effector process| |regulation of response to biotic stimulus| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |protein catabolic process| |protein ubiquitination| |regulation of cytokine production| |viral process| |regulation of defense response| |protein modification by small protein conjugation| |regulation of multi-organism process| |symbiotic process| |interspecies interaction between organisms| |cellular macromolecule catabolic process| |apoptotic process| |protein modification by small protein conjugation or removal| |macromolecule catabolic process| |programmed cell death| |organonitrogen compound catabolic process| |cell death| |regulation of response to external stimulus| |negative regulation of multicellular organismal process| |proteolysis| |regulation of response to stress| |regulation of immune system process| |organic substance catabolic process| |cellular catabolic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |PNU96415E 10μM R03 exp145|-2.32| |IU1-47 25μM R04 exp182|-1.96| |Quinacrine 2.5μM R03 exp146|-1.93| |Epothilone-D 0.004 to 0.006μM on day4 R05 exp232|-1.78| |HMS-I1 1μM R01 exp46|-1.77| |Taxol 0.002μM R01 exp54|-1.75| |Docetaxel 0.002μM R01 exp45|-1.73| |Centrinone 0.5μM R07 exp343|1.87| |VER-155008 3.9μM R06 exp301|2.02| |HMS-I1 10μM R06 exp256|2.1| |SB743921 0.001μM R05 exp244|2.66| |Rotenone 2μM R00 exp33|3| |DABN racemic mixture R03 exp93|3.03| |R-DABN 8μM R03 exp111|3.3| |Nocodazole 0.1μM R03 exp94|3.39| |Nocodazole 0.2μM R00 exp24|3.61| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 10613 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.9 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='RNF216 Expression in NALM6 Cells: 6.9'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
