This protein is involved in cargo-specific control of clathrin-mediated endocytosis, specifically controlling the internalization of a specific protein receptor. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: May function in transferrin receptor internalization at the plasma membrane through a cargo-specific control of clathrin- mediated endocytosis. Alternatively, may act as a negative regulator of the amino acid-induced TOR signaling by inhibiting the formation of active Rag GTPase complexes. Preferentially binds inactive Rag GTPase complexes and prevents their interaction with the mTORC1 complex inhibiting its relocalization to lysosomes and its activation. Thereby, may indirectly regulate cell growth, proliferation and autophagy. {ECO:0000269|PubMed:16325581, ECO:0000269|PubMed:22575674}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |SH3 1| |SH3 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |GDP-dissociation inhibitor activity| |Ras GTPase binding| |amino acid import| |protein localization to lysosome| |negative regulation of GTPase activity| |negative regulation of TOR signaling| |clathrin-coated pit| |clathrin-coated vesicle| |protein localization to vacuole| |cellular response to amino acid stimulus| |regulation of TOR signaling| |response to amino acid| |amino acid transport| |positive regulation of autophagy| |negative regulation of cell growth| |cellular response to acid chemical| |negative regulation of growth| |carboxylic acid transport| |organic acid transport| |regulation of autophagy| |response to acid chemical| |positive regulation of cellular catabolic process| |regulation of cell growth| |positive regulation of catabolic process| |negative regulation of hydrolase activity| |organic anion transport| |regulation of GTPase activity| |negative regulation of intracellular signal transduction| |endocytosis| |anion transport| |cellular response to organonitrogen compound| |cellular response to nitrogen compound| |regulation of growth| |import into cell| |negative regulation of cell population proliferation| |protein localization to organelle| |negative regulation of catalytic activity| |regulation of cellular catabolic process| |regulation of catabolic process| |response to organonitrogen compound| |cellular response to oxygen-containing compound| |identical protein binding| |response to nitrogen compound| |negative regulation of molecular function| |cellular response to endogenous stimulus| |negative regulation of signal transduction| |regulation of hydrolase activity| |negative regulation of cell communication| |negative regulation of signaling| |ion transport| |response to endogenous stimulus| |response to oxygen-containing compound| |cellular protein localization| |cellular macromolecule localization| |regulation of cell population proliferation| |negative regulation of response to stimulus| |regulation of intracellular signal transduction| |nitrogen compound transport| |vesicle-mediated transport| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 8410 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -5.6 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='SH3BP4 Expression in NALM6 Cells: -5.6'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
