Involved in cellular amino acid uptake. Acts as an amino acid exchanger. Involved in the transport of L-DOPA across the blood- brain barrier, and that of thyroid hormones triiodothyronine (T3) and thyroxine (T4) across the cell membrane in tissues such as placenta. Plays a role in neuronal cell proliferation (neurogenesis) in brain. Involved in the uptake of methylmercury (MeHg) when administered as the L-cysteine or D,L-homocysteine complexes, and hence plays a role in metal ion homeostasis and toxicity. Involved in the cellular activity of small molecular weight nitrosothiols, via the stereoselective transport of L- nitrosocysteine (L-CNSO) across the transmembrane. May play an important role in high-grade gliomas. Mediates blood-to-retina L- leucine transport across the inner blood-retinal barrier which in turn may play a key role in maintaining large neutral amino acids as well as neurotransmitters in the neural retina. Acts as the major transporter of tyrosine in fibroblasts. When associated with LAPTM4B, recruits SLC3A2 and SLC7A5 to lysosomes to promote leucine uptake into these organelles and is required for mTORC1 activation (PubMed:25998567). {ECO:0000269|PubMed:10049700, ECO:0000269|PubMed:10391915, ECO:0000269|PubMed:10574970, ECO:0000269|PubMed:11311135, ECO:0000269|PubMed:11389679, ECO:0000269|PubMed:11557028, ECO:0000269|PubMed:11564694, ECO:0000269|PubMed:11742812, ECO:0000269|PubMed:12117417, ECO:0000269|PubMed:12225859, ECO:0000269|PubMed:15769744, ECO:0000269|PubMed:16496379, ECO:0000269|PubMed:18262359, ECO:0000269|PubMed:25998567, ECO:0000269|PubMed:9751058}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |AA permease| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |L-tryptophan transmembrane transport| |amino acid transport complex| |L-tryptophan transmembrane transporter activity| |L-leucine transmembrane transporter activity| |tryptophan transport| |branched-chain amino acid transport| |aromatic amino acid transport| |L-amino acid transmembrane transporter activity| |peptide antigen binding| |amino acid transmembrane transporter activity| |neutral amino acid transport| |L-alpha-amino acid transmembrane transport| |L-amino acid transport| |drug transmembrane transport| |amino acid transmembrane transport| |amino acid transport| |organic acid transmembrane transport| |carboxylic acid transmembrane transport| |drug transport| |anion transmembrane transport| |organic acid transport| |carboxylic acid transport| |apical plasma membrane| |leukocyte migration| |organic anion transport| |anion transport| |cation transmembrane transport| |intracellular membrane-bounded organelle| |cation transport| |ion transmembrane transport| |cell migration| |response to drug| |localization of cell| |cell motility| |transmembrane transport| |locomotion| |ion transport| |movement of cell or subcellular component| |nitrogen compound transport| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Epigallocatechin gallate 20μM R05 exp230|-3.55| |IWR1 50μM R08 exp495|-3.27| |A-196 10μM R03 exp114|-3.17| |SGC2043 10μM R03 exp152|-2.69| |Lapatinib 5μM R01 exp47|-2.45| |Trametinib 10μM R02 exp83|-2.41| |SB203580 25μM R03 exp149|-2.29| |TRAIL 5ng/ml R00 exp35|1.74| |Dimethyloxaloylglycine 100μM R05 exp229|1.77| |Rucaparib 6.5μM R08 exp520|1.86| |DABN 20μM R00 exp17|1.87| |UM0125461 0.74μM R02 exp84|1.92| |Dynasore 7μM R08 exp436|1.93| |CoCl2 18μM R07 exp346|2.05| |Sulforaphane 9μM R08 exp525|2.1| |Tubacin 1.6μM R07 exp416|2.45| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |MIOS|0.419| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 74/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|8/28| |blood|2/28| |bone|3/26| |breast|5/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|2/13| |fibroblast|0/1| |gastric|3/16| |kidney|6/21| |liver|1/20| |lung|7/75| |lymphocyte|4/16| |ovary|1/26| |pancreas|4/24| |peripheral nervous system|0/16| |plasma cell|4/15| |prostate|0/1| |skin|4/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|1/22| |urinary tract|1/29| |uterus|1/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 10497 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 9.36 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='SLC7A5 Expression in NALM6 Cells: 9.36'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
