The FACT complex is involved in multiple processes that require DNA as a template such as mRNA elongation, DNA replication and DNA repair. During transcription elongation the FACT complex acts as a histone chaperone that both destabilizes and restores nucleosomal structure. It facilitates the passage of RNA polymerase II and transcription by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, then subsequently promotes the reestablishment of the nucleosome following the passage of RNA polymerase II. The FACT complex is probably also involved in phosphorylation of 'Ser-392' of p53/TP53 via its association with CK2 (casein kinase II). Binds specifically to double-stranded DNA and at low levels to DNA modified by the antitumor agent cisplatin. May potentiate cisplatin-induced cell death by blocking replication and repair of modified DNA. Also acts as a transcriptional coactivator for p63/TP63. {ECO:0000269|PubMed:10912001, ECO:0000269|PubMed:11239457, ECO:0000269|PubMed:12374749, ECO:0000269|PubMed:12934006, ECO:0000269|PubMed:16713563, ECO:0000269|PubMed:9489704, ECO:0000269|PubMed:9566881, ECO:0000269|PubMed:9836642}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |SSrecog| |Rtt106| |HMG box| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |FACT complex| |nucleosome binding| |transcription elongation from RNA polymerase II promoter| |DNA-templated transcription, elongation| |histone binding| |regulation of signal transduction by p53 class mediator| |DNA replication| |nuclear chromatin| |transcription by RNA polymerase II| |DNA repair| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |DNA metabolic process| |cellular response to DNA damage stimulus| |nucleolus| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |organic cyclic compound biosynthetic process| |RNA binding| |DNA binding| |DNA-binding transcription factor activity, RNA polymerase II-specific| |cellular nitrogen compound biosynthetic process| |RNA metabolic process| |cellular response to stress| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |regulation of intracellular signal transduction| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |NFN1 0.4μM R03 exp99|1.76| |Golgicide-A 4μM R03 exp122|1.84| |NFN1 0.1μM R01 exp49|1.86| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |POLR2J3|0.425| |POLA2|0.423| |AKAP1|0.417| |CBWD3|0.412| |PSMD11|0.409| |ANKLE2|0.404| |MYB|0.401| |SMG1|0.401| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 682/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|0/1| |bile duct|27/28| |blood|26/28| |bone|23/25| |breast|30/33| |central nervous system|53/56| |cervix|4/4| |colorectal|17/17| |esophagus|13/13| |fibroblast|1/1| |gastric|15/15| |kidney|20/21| |liver|18/20| |lung|67/75| |lymphocyte|14/14| |ovary|25/26| |pancreas|22/24| |peripheral nervous system|16/16| |plasma cell|14/15| |prostate|1/1| |skin|19/24| |soft tissue|5/7| |thyroid|2/2| |upper aerodigestive|19/22| |urinary tract|29/29| |uterus|5/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 528 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 8.65 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='SSRP1 Expression in NALM6 Cells: 8.65'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
