Required for the normal activation of the FA pathway, leading to monoubiquitination of the FANCI-FANCD2 complex in response to DNA damage, cellular resistance to DNA cross-linking drugs, and prevention of chromosomal breakage (PubMed:20347428, PubMed:20347429). In complex with CENPS (MHF heterodimer), crucial cofactor for FANCM in both binding and ATP-dependent remodeling of DNA. Stabilizes FANCM. In complex with CENPS and FANCM (but not other FANC proteins), rapidly recruited to blocked forks and promotes gene conversion at blocked replication forks (PubMed:20347428, PubMed:20347429). In complex with CENPS, CENPT and CENPW (CENP-T-W-S-X heterotetramer), involved in the formation of a functional kinetochore outer plate, which is essential for kinetochore-microtubule attachment and faithful mitotic progression (PubMed:19620631). As a component of MHF and CENP-T-W- S-X complexes, binds DNA and bends it to form a nucleosome-like structure (PubMed:20347428, PubMed:20347429). DNA-binding function is fulfilled in the presence of CENPS, with the following preference for DNA substates: Holliday junction > double-stranded > splay arm > single-stranded. Does not bind DNA on its own (PubMed:20347429). {ECO:0000269|PubMed:19620631, ECO:0000269|PubMed:20347428, ECO:0000269|PubMed:20347429}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |CENP-X| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |FANCM-MHF complex| |kinetochore assembly| |Fanconi anaemia nuclear complex| |resolution of meiotic recombination intermediates| |kinetochore organization| |meiotic chromosome separation| |CENP-A containing nucleosome assembly| |chromosome separation| |replication fork processing| |CENP-A containing chromatin organization| |chromatin remodeling at centromere| |centromere complex assembly| |DNA-dependent DNA replication maintenance of fidelity| |DNA replication-independent nucleosome assembly| |DNA replication-independent nucleosome organization| |histone exchange| |reciprocal meiotic recombination| |interstrand cross-link repair| |homologous recombination| |ATP-dependent chromatin remodeling| |meiotic chromosome segregation| |condensed chromosome kinetochore| |double-stranded DNA binding| |meiosis I| |meiosis I cell cycle process| |positive regulation of protein ubiquitination| |nucleosome assembly| |DNA-dependent DNA replication| |positive regulation of protein modification by small protein conjugation or removal| |chromatin assembly| |meiotic nuclear division| |chromatin assembly or disassembly| |nucleosome organization| |chromatin remodeling| |meiotic cell cycle process| |DNA packaging| |regulation of protein ubiquitination| |protein-DNA complex assembly| |DNA replication| |nuclear chromosome segregation| |DNA recombination| |meiotic cell cycle| |regulation of protein modification by small protein conjugation or removal| |protein-DNA complex subunit organization| |chromosome segregation| |nuclear division| |DNA conformation change| |organelle fission| |cell division| |DNA repair| |chromatin organization| |DNA metabolic process| |organelle assembly| |cellular response to DNA damage stimulus| |cellular protein-containing complex assembly| |cell cycle process| |chromosome organization| |positive regulation of protein modification process| |cell cycle| |reproductive process| |reproduction| |DNA binding| |protein-containing complex assembly| |positive regulation of cellular protein metabolic process| |cellular response to stress| |positive regulation of protein metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |regulation of protein modification process| |protein-containing complex subunit organization| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Campthothecin 0.001μM R06 exp270|-3.29| |Olaparib 4μM R08 exp512|-2.5| |BI-6727 0.0125μM R03 exp97|-2.02| |Talazoparib 0.006μM R06 exp299|-1.76| |UMK57 1μM R01 exp59|-1.74| |Rucaparib 6.5μM R08 exp520|-1.72| |Nutlin-3A 1.6μM R06 exp294|3.69| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |APITD1|0.583| |FANCM|0.507| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 4/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|2/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|1/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 2697 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 4.11 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='STRA13 Expression in NALM6 Cells: 4.11'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
