The protein that coordinates these activities is transcription factor IID (TFIID), which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TAFs may participate in basal transcription, serve as coactivators, function in promoter recognition or modify general transcription factors (GTFs) to facilitate complex assembly and transcription initiation. This gene encodes one of the smaller subunits of TFIID that binds weakly to TBP but strongly to TAF1, the largest subunit of TFIID. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2010]. * **<color #00a2e8>UniProt Summary</color>**: TAFs are components of the transcription factor IID (TFIID) complex, PCAF histone acetylase complex and TBP-free TAFII complex (TFTC). TIIFD is multimeric protein complex that plays a central role in mediating promoter responses to various activators and repressors. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |TAF| |DUF1546| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |SLIK (SAGA-like) complex| |aryl hydrocarbon receptor binding| |RNA polymerase II preinitiation complex assembly| |transcription factor TFTC complex| |SAGA complex| |transcription preinitiation complex assembly| |MLL1 complex| |transcription factor TFIID complex| |snRNA transcription by RNA polymerase II| |snRNA transcription| |ncRNA transcription| |transcription initiation from RNA polymerase II promoter| |regulation of signal transduction by p53 class mediator| |protein-DNA complex assembly| |DNA-templated transcription, initiation| |protein-DNA complex subunit organization| |transcription by RNA polymerase II| |protein heterodimerization activity| |negative regulation of cell cycle| |protein-containing complex| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |DNA-binding transcription factor activity| |chromatin organization| |cellular protein-containing complex assembly| |apoptotic process| |programmed cell death| |chromosome organization| |cell death| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |regulation of cell cycle| |organic cyclic compound biosynthetic process| |DNA binding| |protein-containing complex assembly| |cellular nitrogen compound biosynthetic process| |RNA metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |regulation of intracellular signal transduction| |protein-containing complex subunit organization| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Pimelic-diphenylamide-106 5μM R00 exp28|-2.07| |42°C R08 exp539|-1.97| |UM0125461 0.74μM R02 exp84|-1.94| |UM0011500 10μM R05 exp246|-1.7| |Nifuroxazide 5μM R03 exp102|1.84| |Deguelin 0.15μM R05 exp209|1.85| |Geldanamycin 0.01μM R05 exp235|1.97| |Benzoate 20000μM R08 exp456|2.17| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |BRD2|0.432| |POLR2J3|0.401| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 715/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|28/28| |blood|28/28| |bone|25/25| |breast|32/33| |central nervous system|55/56| |cervix|4/4| |colorectal|16/17| |esophagus|13/13| |fibroblast|1/1| |gastric|15/15| |kidney|21/21| |liver|20/20| |lung|74/75| |lymphocyte|14/14| |ovary|26/26| |pancreas|23/24| |peripheral nervous system|16/16| |plasma cell|14/15| |prostate|0/1| |skin|24/24| |soft tissue|7/7| |thyroid|2/2| |upper aerodigestive|22/22| |urinary tract|27/29| |uterus|5/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 696 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.48 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='TAF6 Expression in NALM6 Cells: 5.48'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
