The coagulation process initiates with the formation of a factor VIIa-TF complex, which proteolytically activates additional proteases (factors IX and X) and ultimately leads to the formation of a fibrin clot. The product of this gene inhibits the activated factor X and VIIa-TF proteases in an autoregulatory loop. Inhibition of the encoded protein restores hemostasis in animal models of hemophilia. This gene encodes multiple protein isoforms that differ in their inhibitory activity, specificity and cellular localization. [provided by RefSeq, Jul 2016]. * **<color #00a2e8>UniProt Summary</color>**: Inhibits factor X (X(a)) directly and, in a Xa-dependent way, inhibits VIIa/tissue factor activity, presumably by forming a quaternary Xa/LACI/VIIa/TF complex. It possesses an antithrombotic action and also the ability to associate with lipoproteins in plasma. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Kunitz BPTI| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |blood coagulation, extrinsic pathway| |organelle membrane| |protein activation cascade| |blood coagulation, fibrin clot formation| |endopeptidase inhibitor activity| |negative regulation of blood coagulation| |negative regulation of hemostasis| |negative regulation of coagulation| |negative regulation of wound healing| |caveola| |regulation of blood coagulation| |regulation of hemostasis| |regulation of coagulation| |negative regulation of response to wounding| |serine-type endopeptidase inhibitor activity| |anchored component of membrane| |response to estradiol| |regulation of wound healing| |regulation of response to wounding| |cellular response to interleukin-1| |cellular response to lipopolysaccharide| |cellular response to steroid hormone stimulus| |cellular response to molecule of bacterial origin| |response to interleukin-1| |cellular response to biotic stimulus| |negative regulation of endopeptidase activity| |negative regulation of peptidase activity| |blood coagulation| |coagulation| |hemostasis| |response to lipopolysaccharide| |response to molecule of bacterial origin| |response to steroid hormone| |negative regulation of proteolysis| |negative regulation of response to external stimulus| |regulation of endopeptidase activity| |regulation of peptidase activity| |negative regulation of hydrolase activity| |wound healing| |regulation of body fluid levels| |cellular response to lipid| |cellular response to organic cyclic compound| |response to wounding| |cellular response to hormone stimulus| |cell surface| |response to bacterium| |regulation of proteolysis| |negative regulation of catalytic activity| |response to lipid| |response to hormone| |response to organic cyclic compound| |cellular response to cytokine stimulus| |endoplasmic reticulum| |negative regulation of cellular protein metabolic process| |cellular response to oxygen-containing compound| |regulation of response to external stimulus| |response to cytokine| |negative regulation of protein metabolic process| |negative regulation of molecular function| |negative regulation of multicellular organismal process| |cellular response to endogenous stimulus| |regulation of hydrolase activity| |response to other organism| |response to external biotic stimulus| |response to biotic stimulus| |response to endogenous stimulus| |regulation of response to stress| |response to oxygen-containing compound| |extracellular space| |negative regulation of response to stimulus| |extracellular region| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp52|Ribavirin 10μM R01 exp52]]|-2.01| |[[:results:exp282|Fluvastatin 2.2μM R06 exp282]]|-1.73| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 11043 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.01 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='TFPI Expression in NALM6 Cells: 6.01'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
