This binding is mediated by a unique sequence of 47 amino acids C-terminal to an SH3 domain. The protein may be involved in a regulatory mechanism that sustains the GTP-bound active form of Cdc42Hs and which is directly linked to a tyrosine phosphorylation signal transduction pathway. Several alternatively spliced transcript variants have been identified from this gene, but the full-length nature of only two transcript variants has been determined. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Non-receptor tyrosine-protein and serine/threonine- protein kinase that is implicated in cell spreading and migration, cell survival, cell growth and proliferation. Transduces extracellular signals to cytosolic and nuclear effectors. Phosphorylates AKT1, AR, MCF2, WASL and WWOX. Implicated in trafficking and clathrin-mediated endocytosis through binding to epidermal growth factor receptor (EGFR) and clathrin. Binds to both poly- and mono-ubiquitin and regulates ligand-induced degradation of EGFR, thereby contributing to the accumulation of EGFR at the limiting membrane of early endosomes. Downstream effector of CDC42 which mediates CDC42-dependent cell migration via phosphorylation of BCAR1. May be involved both in adult synaptic function and plasticity and in brain development. Activates AKT1 by phosphorylating it on 'Tyr-176'. Phosphorylates AR on 'Tyr-267' and 'Tyr-363' thereby promoting its recruitment to androgen-responsive enhancers (AREs). Phosphorylates WWOX on 'Tyr- 287'. Phosphorylates MCF2, thereby enhancing its activity as a guanine nucleotide exchange factor (GEF) toward Rho family proteins. Contributes to the control of AXL receptor levels. Confers metastatic properties on cancer cells and promotes tumor growth by negatively regulating tumor suppressor such as WWOX and positively regulating pro-survival factors such as AKT1 and AR. Phosphorylates WASP (PubMed:20110370). {ECO:0000269|PubMed:10652228, ECO:0000269|PubMed:11278436, ECO:0000269|PubMed:16247015, ECO:0000269|PubMed:16257963, ECO:0000269|PubMed:16472662, ECO:0000269|PubMed:17038317, ECO:0000269|PubMed:18262180, ECO:0000269|PubMed:18435854, ECO:0000269|PubMed:19815557, ECO:0000269|PubMed:20110370, ECO:0000269|PubMed:20333297, ECO:0000269|PubMed:20383201}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Pkinase| |Pkinase Tyr| |GTPase binding| |Inhibitor Mig-6| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |Grb2-EGFR complex| |cytoophidium| |GTPase inhibitor activity| |regulation of clathrin-dependent endocytosis| |protein serine/threonine/tyrosine kinase activity| |WW domain binding| |peptidyl-tyrosine autophosphorylation| |epidermal growth factor receptor binding| |non-membrane spanning protein tyrosine kinase activity| |negative regulation of GTPase activity| |clathrin-coated pit| |clathrin-coated vesicle| |adherens junction| |protein tyrosine kinase activity| |regulation of receptor-mediated endocytosis| |cytoplasmic vesicle membrane| |peptidyl-tyrosine phosphorylation| |peptidyl-tyrosine modification| |positive regulation of peptidyl-tyrosine phosphorylation| |protein autophosphorylation| |regulation of endocytosis| |regulation of peptidyl-tyrosine phosphorylation| |endosome| |ubiquitin protein ligase binding| |small GTPase mediated signal transduction| |protein serine/threonine kinase activity| |negative regulation of hydrolase activity| |regulation of GTPase activity| |regulation of vesicle-mediated transport| |endocytosis| |import into cell| |perinuclear region of cytoplasm| |negative regulation of catalytic activity| |peptidyl-amino acid modification| |protein phosphorylation| |positive regulation of protein phosphorylation| |positive regulation of phosphorylation| |identical protein binding| |positive regulation of phosphorus metabolic process| |positive regulation of phosphate metabolic process| |negative regulation of molecular function| |positive regulation of protein modification process| |regulation of hydrolase activity| |phosphorylation| |regulation of protein phosphorylation| |ATP binding| |regulation of phosphorylation| |positive regulation of cellular protein metabolic process| |regulation of cell population proliferation| |intracellular signal transduction| |positive regulation of protein metabolic process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| |regulation of transport| |vesicle-mediated transport| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp234|Ethanol 0.01 R05 exp234]]|1.76| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5510 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.87 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='TNK2 Expression in NALM6 Cells: 5.87'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
