Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. Several tryptase genes are clustered on chromosome 16p13.3. These genes are characterized by several distinct features. They have a highly conserved 3' UTR and contain tandem repeat sequences at the 5' flank and 3' UTR which are thought to play a role in regulation of the mRNA stability. These genes have an intron immediately upstream of the initiator Met codon, which separates the site of transcription initiation from protein coding sequence. This feature is characteristic of tryptases but is unusual in other genes. The alleles of this gene exhibit an unusual amount of sequence variation, such that the alleles were once thought to represent two separate genes, beta II and beta III. Beta tryptases appear to be the main isoenzymes expressed in mast cells, whereas in basophils, alpha-tryptases predominate. Tryptases have been implicated as mediators in the pathogenesis of asthma and other allergic and inflammatory disorders. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Tryptase is the major neutral protease present in mast cells and is secreted upon the coupled activation-degranulation response of this cell type. May play a role in innate immunity. {ECO:0000250, ECO:0000250|UniProtKB:P21845}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Trypsin| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |serine-type peptidase activity| |serine-type endopeptidase activity| |collagen-containing extracellular matrix| |proteolysis| |extracellular space| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Hydroxyurea 100μM R07 exp329|-1.94| |BML-284 0.09μM R08 exp460|-1.87| |ABT-702 5μM R07 exp318|-1.73| |Bleomycin 5μM R08 exp459|-1.73| |Daidzin 10μM R06 exp280|1.74| |L-BMAA 500 to 750μM on day4 R04 exp185|1.74| |IU1-C 25μM R04 exp183|2.38| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |HGC6.3|0.511| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: N/A ^Tissue^Fraction Of Cell Lines Where Essential^ </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 6595 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -0.12 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='TPSB2 Expression in NALM6 Cells: -0.12'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
