This gene functionally complements Chinese hamster irs1SF, a repair-deficient mutant that exhibits hypersensitivity to a number of different DNA-damaging agents and is chromosomally unstable. A rare microsatellite polymorphism in this gene is associated with cancer in patients of varying radiosensitivity. Alternatively spliced transcript variants encoding the same protein have been identified. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Involved in the homologous recombination repair (HRR) pathway of double-stranded DNA, thought to repair chromosomal fragmentation, translocations and deletions. Part of the RAD21 paralog protein complex CX3 which acts in the BRCA1-BRCA2- dependent HR pathway. Upon DNA damage, CX3 acts downstream of RAD51 recruitment; the complex binds predominantly to the intersection of the four duplex arms of the Holliday junction (HJ) and to junctions of replication forks. Involved in HJ resolution and thus in processing HR intermediates late in the DNA repair process; the function may be linked to the CX3 complex and seems to involve GEN1 during mitotic cell cycle progression. Part of a PALB2-scaffolded HR complex containing BRCA2 and RAD51C and which is thought to play a role in DNA repair by HR. Plays a role in regulating mitochondrial DNA copy number under conditions of oxidative stress in the presence of RAD51 and RAD51C. {ECO:0000269|PubMed:14716019, ECO:0000269|PubMed:20413593, ECO:0000269|PubMed:23108668, ECO:0000269|PubMed:23149936}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Rad51| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |resolution of mitotic recombination intermediates| |Rad51C-XRCC3 complex| |resolution of recombination intermediates| |positive regulation of spindle checkpoint| |positive regulation of mitotic cell cycle spindle assembly checkpoint| |double-strand break repair via synthesis-dependent strand annealing| |positive regulation of cell cycle checkpoint| |crossover junction endodeoxyribonuclease activity| |telomere maintenance via telomere trimming| |t-circle formation| |formation of extrachromosomal circular DNA| |telomeric loop disassembly| |telomere maintenance via recombination| |regulation of mitotic spindle checkpoint| |regulation of spindle checkpoint| |regulation of mitotic cell cycle spindle assembly checkpoint| |four-way junction DNA binding| |mitotic recombination| |replication fork| |regulation of cell cycle checkpoint| |regulation of centrosome duplication| |DNA-dependent ATPase activity| |regulation of mitotic metaphase/anaphase transition| |interstrand cross-link repair| |regulation of metaphase/anaphase transition of cell cycle| |regulation of mitotic sister chromatid separation| |regulation of centrosome cycle| |regulation of chromosome separation| |regulation of mitotic sister chromatid segregation| |regulation of sister chromatid segregation| |double-strand break repair via homologous recombination| |telomere maintenance| |recombinational repair| |telomere organization| |regulation of chromosome segregation| |nuclear chromosome, telomeric region| |positive regulation of mitotic cell cycle| |regulation of mitotic nuclear division| |double-strand break repair| |regulation of microtubule cytoskeleton organization| |regulation of nuclear division| |DNA recombination| |regulation of microtubule-based process| |positive regulation of cell cycle process| |anatomical structure homeostasis| |regulation of chromosome organization| |positive regulation of cell cycle| |regulation of mitotic cell cycle phase transition| |regulation of cell cycle phase transition| |DNA repair| |regulation of cytoskeleton organization| |regulation of mitotic cell cycle| |perinuclear region of cytoplasm| |DNA metabolic process| |regulation of cell cycle process| |cellular response to DNA damage stimulus| |chromosome organization| |regulation of cell cycle| |mitochondrion| |regulation of organelle organization| |ATP binding| |homeostatic process| |cellular response to stress| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |THZ531 0.225μM R08 exp444|-2.02| |Olaparib 4μM R08 exp512|-1.82| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |XRCC2|0.457| |ATXN10|0.431| |RTCB|0.429| |GMPPB|0.417| |ADSL|0.41| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 238/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|0/1| |bile duct|8/28| |blood|12/28| |bone|9/26| |breast|11/33| |central nervous system|19/56| |cervix|0/4| |colorectal|3/17| |esophagus|5/13| |fibroblast|0/1| |gastric|4/16| |kidney|7/21| |liver|3/20| |lung|17/75| |lymphocyte|2/16| |ovary|6/26| |pancreas|10/24| |peripheral nervous system|2/16| |plasma cell|8/15| |prostate|0/1| |skin|8/24| |soft tissue|3/9| |thyroid|1/2| |upper aerodigestive|10/22| |urinary tract|12/29| |uterus|1/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 150 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.05 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='XRCC3 Expression in NALM6 Cells: 6.05'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
