ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the White subfamily. The protein encoded by this gene functions as a half-transporter to limit intestinal absorption and promote biliary excretion of sterols. It is expressed in a tissue-specific manner in the liver, colon, and intestine. This gene is tandemly arrayed on chromosome 2, in a head-to-head orientation with family member ABCG8. Mutations in this gene may contribute to sterol accumulation and atheroschlerosis, and have been observed in patients with sitosterolemia. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: ABCG5 and ABCG8 form an obligate heterodimer that mediates Mg(2+)- and ATP-dependent sterol transport across the cell membrane (PubMed:27144356). Plays an essential role in the selective transport of dietary plant sterols and cholesterol in and out of the enterocytes and in the selective sterol excretion by the liver into bile (PubMed:11099417, PubMed:11138003, PubMed:27144356, PubMed:15054092). Required for normal sterol homeostasis (PubMed:11099417, PubMed:11138003, PubMed:15054092). The heterodimer with ABCG8 has ATPase activity (PubMed:16893193, PubMed:20210363, PubMed:27144356). {ECO:0000269|PubMed:11138003, ECO:0000269|PubMed:15054092, ECO:0000269|PubMed:16893193, ECO:0000269|PubMed:20210363, ECO:0000269|PubMed:27144356, ECO:0000303|PubMed:11590207, ECO:0000305|PubMed:11099417}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |ABC2 membrane| |ABC tran| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |negative regulation of intestinal cholesterol absorption| |negative regulation of intestinal phytosterol absorption| |negative regulation of intestinal lipid absorption| |negative regulation of intestinal absorption| |ATP-binding cassette (ABC) transporter complex| |regulation of intestinal cholesterol absorption| |regulation of intestinal lipid absorption| |regulation of intestinal absorption| |intestinal cholesterol absorption| |negative regulation of cholesterol transport| |negative regulation of sterol transport| |intestinal lipid absorption| |lipid digestion| |cholesterol transfer activity| |negative regulation of digestive system process| |cholesterol efflux| |negative regulation of lipid transport| |intestinal absorption| |regulation of digestive system process| |excretion| |negative regulation of lipid localization| |regulation of cholesterol transport| |regulation of sterol transport| |ATPase-coupled transmembrane transporter activity| |cholesterol transport| |sterol transport| |digestive system process| |cholesterol homeostasis| |sterol homeostasis| |regulation of lipid transport| |digestion| |lipid homeostasis| |organic hydroxy compound transport| |regulation of lipid localization| |response to ionizing radiation| |receptor complex| |response to nutrient| |ATPase activity| |lipid transport| |apical plasma membrane| |lipid localization| |response to radiation| |negative regulation of transport| |protein heterodimerization activity| |response to nutrient levels| |response to extracellular stimulus| |regulation of system process| |ion transmembrane transport| |response to drug| |chemical homeostasis| |response to abiotic stimulus| |negative regulation of multicellular organismal process| |transmembrane transport| |ion transport| |integral component of plasma membrane| |ATP binding| |homeostatic process| |regulation of transport| |system process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Lenalidomide 20μM R07 exp375|-2.56| |Genistein 15μM R07 exp360|-1.75| |Citral 75μM R06 exp275|-1.74| |Daidzin 10μM R06 exp280|-1.7| |Glyphosate 1000μM R06 exp283|1.79| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5673 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -0.36 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='ABCG5 Expression in NALM6 Cells: -0.36'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
