Required for the normal activation of the FA pathway, leading to monoubiquitination of the FANCI-FANCD2 complex in response to DNA damage, cellular resistance to DNA cross-linking drugs, and prevention of chromosomal breakage (PubMed:20347428, PubMed:20347429). In complex with CENPX (MHF heterodimer), crucial cofactor for FANCM in both binding and ATP-dependent remodeling of DNA. Stabilizes FANCM (PubMed:20347428, PubMed:20347429). In complex with CENPX and FANCM (but not other FANC proteins), rapidly recruited to blocked forks and promotes gene conversion at blocked replication forks (PubMed:20347428). In complex with CENPT, CENPW and CENPX (CENP-T-W-S-X heterotetramer), involved in the formation of a functional kinetochore outer plate, which is essential for kinetochore-microtubule attachment and faithful mitotic progression (PubMed:19620631). As a component of MHF and CENP-T-W-S-X complexes, binds DNA and bends it to form a nucleosome-like structure (PubMed:20347428, PubMed:22304917). DNA- binding function is fulfilled in the presence of CENPX, with the following preference for DNA substates: Holliday junction > double-stranded > splay arm > single-stranded. Does not bind DNA on its own (PubMed:20347428, PubMed:20347429). {ECO:0000269|PubMed:19620631, ECO:0000269|PubMed:20347428, ECO:0000269|PubMed:20347429, ECO:0000269|PubMed:22304917}. <button type='default' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> No Pfam Domain information is available for this gene. </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |FANCM-MHF complex| |kinetochore assembly| |Fanconi anaemia nuclear complex| |resolution of meiotic recombination intermediates| |kinetochore organization| |meiotic chromosome separation| |CENP-A containing nucleosome assembly| |chromosome separation| |replication fork processing| |CENP-A containing chromatin organization| |chromatin remodeling at centromere| |centromere complex assembly| |DNA replication-independent nucleosome assembly| |DNA-dependent DNA replication maintenance of fidelity| |DNA replication-independent nucleosome organization| |histone exchange| |reciprocal meiotic recombination| |homologous recombination| |interstrand cross-link repair| |ATP-dependent chromatin remodeling| |condensed chromosome kinetochore| |meiotic chromosome segregation| |double-stranded DNA binding| |meiosis I| |meiosis I cell cycle process| |positive regulation of protein ubiquitination| |nucleosome assembly| |DNA-dependent DNA replication| |positive regulation of protein modification by small protein conjugation or removal| |chromatin assembly| |meiotic nuclear division| |chromatin assembly or disassembly| |nucleosome organization| |chromatin remodeling| |meiotic cell cycle process| |DNA packaging| |regulation of protein ubiquitination| |protein-DNA complex assembly| |DNA replication| |nuclear chromosome segregation| |DNA recombination| |meiotic cell cycle| |regulation of protein modification by small protein conjugation or removal| |protein-DNA complex subunit organization| |chromosome segregation| |nuclear division| |DNA conformation change| |organelle fission| |chromatin binding| |protein heterodimerization activity| |cell division| |DNA repair| |chromatin organization| |DNA metabolic process| |organelle assembly| |cellular response to DNA damage stimulus| |cellular protein-containing complex assembly| |cell cycle process| |chromosome organization| |positive regulation of protein modification process| |cell cycle| |reproductive process| |reproduction| |DNA binding| |protein-containing complex assembly| |positive regulation of cellular protein metabolic process| |cellular response to stress| |positive regulation of protein metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |regulation of protein modification process| |protein-containing complex subunit organization| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Olaparib 4μM R08 exp512|-3.7| |Rucaparib 6.5μM R08 exp520|-3.39| |BI-2536 0.0042μM R03 exp95|-2.31| |BI-6727 0.0125μM R03 exp97|-2.01| |Campthothecin 0.001μM R06 exp270|-1.77| |Nutlin-3A 1.6μM R06 exp294|4.79| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |STRA13|0.583| |FANCM|0.403| |SWSAP1|0.4| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 6310 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 3.0 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='APITD1 Expression in NALM6 Cells: 3.0'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
