Recognizes and selectively binds the transmembrane domain of TA proteins in the cytosol. This complex then targets to the endoplasmic reticulum by membrane-bound receptors, where the tail- anchored protein is released for insertion. This process is regulated by ATP binding and hydrolysis. ATP binding drives the homodimer towards the closed dimer state, facilitating recognition of newly synthesized TA membrane proteins. ATP hydrolysis is required for insertion. Subsequently, the homodimer reverts towards the open dimer state, lowering its affinity for the membrane-bound receptor, and returning it to the cytosol to initiate a new round of targeting (By similarity). May be involved in insulin signaling. {ECO:0000255|HAMAP-Rule:MF_03112, ECO:0000269|PubMed:17382883, ECO:0000269|PubMed:18477612}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |ArsA ATPase| |CbiA| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |GET complex| |arsenite transmembrane transporter activity| |arsenite transport| |tail-anchored membrane protein insertion into ER membrane| |protein insertion into ER membrane| |posttranslational protein targeting to endoplasmic reticulum membrane| |protein insertion into membrane| |IRE1-mediated unfolded protein response| |endoplasmic reticulum organization| |endoplasmic reticulum unfolded protein response| |protein targeting to ER| |establishment of protein localization to endoplasmic reticulum| |cellular response to unfolded protein| |protein localization to endoplasmic reticulum| |cellular response to topologically incorrect protein| |inorganic anion transport| |protein targeting to membrane| |response to unfolded protein| |response to topologically incorrect protein| |ATPase activity| |response to endoplasmic reticulum stress| |establishment of protein localization to membrane| |protein targeting| |endomembrane system organization| |establishment of protein localization to organelle| |protein localization to membrane| |anion transport| |protein localization to organelle| |membrane organization| |nucleolus| |endoplasmic reticulum membrane| |intracellular protein transport| |ion transport| |ATP binding| |protein transport| |intracellular transport| |peptide transport| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |cellular response to stress| |establishment of localization in cell| |nitrogen compound transport| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Vemurafenib 6.6μM R02 exp89|-1.76| |5-Fluorouracil 2μM R00 exp1|-1.7| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |GET4|0.429| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 246/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|1/1| |bile duct|14/28| |blood|11/28| |bone|7/26| |breast|10/33| |central nervous system|19/56| |cervix|1/4| |colorectal|9/17| |esophagus|8/13| |fibroblast|0/1| |gastric|3/16| |kidney|14/21| |liver|8/20| |lung|27/75| |lymphocyte|8/16| |ovary|4/26| |pancreas|9/24| |peripheral nervous system|2/16| |plasma cell|7/15| |prostate|1/1| |skin|5/24| |soft tissue|1/9| |thyroid|1/2| |upper aerodigestive|8/22| |urinary tract|13/29| |uterus|1/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 520 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.52 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='ASNA1 Expression in NALM6 Cells: 5.52'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
