The encoded membrane protein represents the delta subunit of the CD3 complex, and along with four other CD3 subunits, binds either TCR alpha/beta or TCR gamma/delta to form the TCR/CD3 complex on the surface of T-cells. Defects in this gene are a cause of severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (SCIDBNK). Two transcript variants encoding different isoforms have been found for this gene. Other variants may also exist, but the full-length natures of their transcripts has yet to be defined. [provided by RefSeq, Feb 2009]. * **<color #00a2e8>UniProt Summary</color>**: Part of the TCR-CD3 complex present on T-lymphocyte cell surface that plays an essential role in adaptive immune response. When antigen presenting cells (APCs) activate T-cell receptor (TCR), TCR-mediated signals are transmitted across the cell membrane by the CD3 chains CD3D, CD3E, CD3G and CD3Z. All CD3 chains contain immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domain. Upon TCR engagement, these motifs become phosphorylated by Src family protein tyrosine kinases LCK and FYN, resulting in the activation of downstream signaling pathways (PubMed:2470098). In addition of this role of signal transduction in T-cell activation, CD3D plays an essential role in thymocyte differentiation. Indeed, participates in correct intracellular TCR-CD3 complex assembly and surface expression. In absence of a functional TCR-CD3 complex, thymocytes are unable to differentiate properly. Interacts with CD4 and CD8 and thus serves to establish a functional link between the TCR and coreceptors CD4 and CD8, which is needed for activation and positive selection of CD4 or CD8 T-cells(PubMed:12215456). {ECO:0000269|PubMed:12215456, ECO:0000269|PubMed:12507424, ECO:0000269|PubMed:2470098}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |ITAM| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |alpha-beta T cell receptor complex| |positive thymic T cell selection| |thymic T cell selection| |positive T cell selection| |T cell selection| |T cell differentiation in thymus| |clathrin-coated vesicle membrane| |T cell receptor complex| |T cell differentiation| |transmembrane signaling receptor activity| |T cell receptor signaling pathway| |T cell activation| |lymphocyte differentiation| |transcription coactivator activity| |antigen receptor-mediated signaling pathway| |leukocyte differentiation| |protein homooligomerization| |lymphocyte activation| |external side of plasma membrane| |immune response-activating cell surface receptor signaling pathway| |immune response-regulating cell surface receptor signaling pathway| |protein heterodimerization activity| |protein complex oligomerization| |hemopoiesis| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |hematopoietic or lymphoid organ development| |adaptive immune response| |activation of immune response| |immune system development| |membrane organization| |positive regulation of immune response| |protein homodimerization activity| |leukocyte activation| |cell activation| |positive regulation of immune system process| |regulation of immune response| |positive regulation of transcription by RNA polymerase II| |positive regulation of transcription, DNA-templated| |protein-containing complex assembly| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |regulation of immune system process| |positive regulation of RNA metabolic process| |protein-containing complex subunit organization| |immune response| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/25| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/15| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/14| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/7| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 16147 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -6.6 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='CD3D Expression in NALM6 Cells: -6.6'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
