The encoded protein is present in all cell types and localized in plasma membranes and in intracellular vesicles. It is a multi-pass membrane protein which contains a ClC domain and two additional C-terminal CBS (cystathionine beta-synthase) domains. The ClC domain catalyzes the selective flow of Cl- ions across cell membranes, and the CBS domain may have a regulatory function. This protein plays a role in both acidification and transmitter loading of GABAergic synaptic vesicles, and in smooth muscle cell activation and neointima formation. This protein is required for lysophosphatidic acid (LPA)-activated Cl- current activity and fibroblast-to-myofibroblast differentiation. The protein activity is regulated by Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) in glioma cells. Multiple alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]. * **<color #00a2e8>UniProt Summary</color>**: Mediates the exchange of chloride ions against protons. Functions as antiporter and contributes to the acidification of the endosome and synaptic vesicle lumen, and may thereby affect vesicle trafficking and exocytosis. May play an important role in neuronal cell function through regulation of membrane excitability by protein kinase C. It could help neuronal cells to establish short-term memory. {ECO:0000269|PubMed:11967229}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Voltage CLC| |CBS| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |synaptic vesicle lumen acidification| |endosomal lumen acidification| |volume-sensitive chloride channel activity| |negative regulation of cell volume| |solute:proton antiporter activity| |voltage-gated chloride channel activity| |synaptic vesicle maturation| |chloride ion binding| |specific granule| |cell volume homeostasis| |vesicle membrane| |neuron cellular homeostasis| |transport vesicle membrane| |intracellular pH reduction| |pH reduction| |chloride channel activity| |phagocytic vesicle| |endosome organization| |regulation of intracellular pH| |secretory granule| |PDZ domain binding| |chloride transmembrane transport| |regulation of cellular pH| |regulation of pH| |chloride transport| |inorganic anion transmembrane transport| |cellular monovalent inorganic cation homeostasis| |synaptic vesicle cycle| |late endosome membrane| |synaptic vesicle| |late endosome| |vesicle-mediated transport in synapse| |monovalent inorganic cation homeostasis| |proton transmembrane transport| |early endosome membrane| |inorganic anion transport| |regulation of cell size| |developmental maturation| |cytoplasmic vesicle| |early endosome| |endosome| |anion transmembrane transport| |vesicle organization| |anatomical structure homeostasis| |regulation of cellular component size| |external side of plasma membrane| |monovalent inorganic cation transport| |endomembrane system organization| |protein heterodimerization activity| |regulation of anatomical structure size| |inorganic cation transmembrane transport| |anion transport| |cation transmembrane transport| |cell surface| |Golgi membrane| |cellular cation homeostasis| |cellular ion homeostasis| |inorganic ion transmembrane transport| |cation homeostasis| |inorganic ion homeostasis| |cellular chemical homeostasis| |ion homeostasis| |cation transport| |protein homodimerization activity| |cellular homeostasis| |ion transmembrane transport| |Golgi apparatus| |chemical homeostasis| |transmembrane transport| |ion transport| |integral component of plasma membrane| |ATP binding| |homeostatic process| |establishment of localization in cell| |vesicle-mediated transport| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp227|Cryptotanshinone 12μM R05 exp227]]|-2.52| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 14160 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.48 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='CLCN3 Expression in NALM6 Cells: 6.48'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
