Participates in the protection of telomeres against non-homologous end-joining (NHEJ)- mediated repair, thereby ensuring that telomeres do not fuse. Plays a key role in telomeric loop (T loop) formation by being recruited by TERF2 at the leading end telomeres and by processing leading-end telomeres immediately after their replication via its exonuclease activity: generates 3' single-stranded overhang at the leading end telomeres avoiding blunt leading-end telomeres that are vulnerable to end-joining reactions and expose the telomere end in a manner that activates the DNA repair pathways. Together with TERF2, required to protect telomeres from replicative damage during replication by controlling the amount of DNA topoisomerase (TOP1, TOP2A and TOP2B) needed for telomere replication during fork passage and prevent aberrant telomere topology. Also involved in response to DNA damage: plays a role in response to DNA interstrand cross-links (ICLs) by facilitating double-strand break formation. In case of spindle stress, involved in prophase checkpoint. {ECO:0000269|PubMed:15467758, ECO:0000269|PubMed:15572677, ECO:0000269|PubMed:16730175, ECO:0000269|PubMed:16730176, ECO:0000269|PubMed:18468965, ECO:0000269|PubMed:18469862, ECO:0000269|PubMed:19197158, ECO:0000269|PubMed:19411856, ECO:0000269|PubMed:20655466}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |DRMBL| |Lactamase B 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |telomeric loop formation| |telomeric 3 overhang formation| |5-3 exodeoxyribonuclease activity| |protection from non-homologous end joining at telomere| |telomere maintenance in response to DNA damage| |5-3 exonuclease activity| |telomere capping| |DNA strand elongation| |telomere maintenance via telomere lengthening| |chromosome, telomeric region| |interstrand cross-link repair| |damaged DNA binding| |double-strand break repair via nonhomologous end joining| |non-recombinational repair| |telomere maintenance| |telomere organization| |nuclear chromosome, telomeric region| |mitotic cell cycle checkpoint| |double-strand break repair| |cell cycle checkpoint| |protein-containing complex binding| |nuclear body| |nucleic acid phosphodiester bond hydrolysis| |negative regulation of mitotic cell cycle| |anatomical structure homeostasis| |centrosome| |DNA repair| |negative regulation of cell cycle| |mitotic cell cycle process| |regulation of mitotic cell cycle| |mitotic cell cycle| |DNA metabolic process| |cellular response to DNA damage stimulus| |protein homodimerization activity| |cell cycle process| |chromosome organization| |regulation of cell cycle| |cell cycle| |homeostatic process| |cellular response to stress| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Nutlin-3A 1.6μM R06 exp294|-2.75| |AICAR 240μM R03 exp116|-2.21| |Mdivi-1 15μM R05 exp217|-2.2| |TRAIL 5ng/ml R00 exp35|-2.19| |Rapamycin 2μM plus FK-506 5μM R07 exp308|-2.14| |Ibrutinib 1μM R07 exp372|-2.1| |Ibrutinib 10μM R08 exp442|-1.91| |NN-Dimethylsphingosine 2.5μM R08 exp508|-1.87| |Tubastatin-A 2.5μM R07 exp417|-1.87| |Hippuristanol 0.12μM R08 exp488|-1.79| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 254/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|12/28| |blood|3/28| |bone|13/26| |breast|10/33| |central nervous system|29/56| |cervix|1/4| |colorectal|8/17| |esophagus|2/13| |fibroblast|1/1| |gastric|5/16| |kidney|7/21| |liver|5/20| |lung|19/75| |lymphocyte|3/16| |ovary|9/26| |pancreas|9/24| |peripheral nervous system|11/16| |plasma cell|1/15| |prostate|0/1| |skin|16/24| |soft tissue|3/9| |thyroid|0/2| |upper aerodigestive|5/22| |urinary tract|6/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1972 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.12 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='DCLRE1B Expression in NALM6 Cells: 5.12'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
