This enzyme hydrolyzes the triphosphate linkage of the cap structure (7-methylguanosine nucleoside triphosphate) to yield 7-methylguanosine monophosphate and nucleoside diphosphate. It protects the cell from the potentially toxic accumulation of these short, capped mRNA fragments, and regulates the activity of other cap-binding proteins, which are inhibited by their accumulation. It also acts as a transcript-specific modulator of pre-mRNA splicing and microRNA turnover. [provided by RefSeq, Apr 2017]. * **<color #00a2e8>UniProt Summary</color>**: Decapping scavenger enzyme that catalyzes the cleavage of a residual cap structure following the degradation of mRNAs by the 3'->5' exosome-mediated mRNA decay pathway. Hydrolyzes cap analog structures like 7-methylguanosine nucleoside triphosphate (m7GpppG) with up to 10 nucleotide substrates (small capped oligoribonucleotides) and specifically releases 5'-phosphorylated RNA fragments and 7-methylguanosine monophosphate (m7GMP). Cleaves cap analog structures like tri-methyl guanosine nucleoside triphosphate (m3(2,2,7)GpppG) with very poor efficiency. Does not hydrolyze unmethylated cap analog (GpppG) and shows no decapping activity on intact m7GpppG-capped mRNA molecules longer than 25 nucleotides. Does not hydrolyze 7-methylguanosine diphosphate (m7GDP) to m7GMP (PubMed:22985415). May also play a role in the 5'->3 mRNA decay pathway; m7GDP, the downstream product released by the 5'->3' mRNA mediated decapping activity, may be also converted by DCPS to m7GMP (PubMed:14523240). Binds to m7GpppG and strongly to m7GDP. Plays a role in first intron splicing of pre- mRNAs. Inhibits activation-induced cell death. {ECO:0000269|PubMed:11747811, ECO:0000269|PubMed:12198172, ECO:0000269|PubMed:12871939, ECO:0000269|PubMed:14523240, ECO:0000269|PubMed:15273322, ECO:0000269|PubMed:15383679, ECO:0000269|PubMed:15769464, ECO:0000269|PubMed:16140270, ECO:0000269|PubMed:18426921, ECO:0000269|PubMed:22985415}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |DcpS C| |DcpS| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |cellular response to menadione| |deadenylation-dependent decapping of nuclear-transcribed mRNA| |m7G(5)pppN diphosphatase activity| |RNA 7-methylguanosine cap binding| |exoribonuclease activity| |mRNA cis splicing, via spliceosome| |exonucleolytic catabolism of deadenylated mRNA| |nuclear-transcribed mRNA catabolic process, exonucleolytic| |RNA phosphodiester bond hydrolysis, exonucleolytic| |nuclear-transcribed mRNA catabolic process, deadenylation-dependent decay| |P-body| |cellular response to ketone| |RNA phosphodiester bond hydrolysis| |response to ketone| |nuclear-transcribed mRNA catabolic process| |mRNA catabolic process| |RNA catabolic process| |RNA splicing, via transesterification reactions with bulged adenosine as nucleophile| |mRNA splicing, via spliceosome| |nucleic acid phosphodiester bond hydrolysis| |RNA splicing, via transesterification reactions| |nucleobase-containing compound catabolic process| |RNA splicing| |cellular response to drug| |heterocycle catabolic process| |cellular nitrogen compound catabolic process| |aromatic compound catabolic process| |mRNA processing| |organic cyclic compound catabolic process| |mRNA metabolic process| |RNA processing| |cellular macromolecule catabolic process| |negative regulation of programmed cell death| |negative regulation of cell death| |response to drug| |macromolecule catabolic process| |cellular response to oxygen-containing compound| |mitochondrion| |response to oxygen-containing compound| |regulation of programmed cell death| |RNA metabolic process| |regulation of cell death| |negative regulation of gene expression| |organic substance catabolic process| |cellular catabolic process| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |IU1-47 25μM R04 exp182|-2.28| |Dexamethasone 0.006μM R07 exp351|-1.78| |Nicotinamide 1000μM R03 exp101|-1.76| |BI-6727 0.001μM R01 exp42|1.8| |2-Deoxy-D-glucose 800μM R05 exp214|2.17| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |TRMT61A|0.451| |PRIM1|0.424| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 11/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|3/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|1/13| |fibroblast|0/1| |gastric|0/16| |kidney|1/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 713 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 3.7 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='DCPS Expression in NALM6 Cells: 3.7'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
