Dynactin binds to both microtubules and cytoplasmic dynein. It is involved in a diverse array of cellular functions, including ER-to-Golgi transport, the centripetal movement of lysosomes and endosomes, spindle formation, chromosome movement, nuclear positioning, and axonogenesis. This subunit is present in 4-5 copies per dynactin molecule. It contains three short alpha-helical coiled-coil domains that may mediate association with self or other dynactin subunits. It may interact directly with the largest subunit (p150) of dynactin and may affix p150 in place. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, May 2012]. * **<color #00a2e8>UniProt Summary</color>**: Modulates cytoplasmic dynein binding to an organelle, and plays a role in prometaphase chromosome alignment and spindle organization during mitosis. Involved in anchoring microtubules to centrosomes. May play a role in synapse formation during brain development. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Dynamitin| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |dynactin complex| |protein localization to microtubule organizing center| |melanosome transport| |protein localization to centrosome| |pigment granule transport| |establishment of melanosome localization| |establishment of pigment granule localization| |melanosome localization| |pigment granule localization| |dynein complex| |spectrin binding| |protein localization to microtubule cytoskeleton| |protein localization to cytoskeleton| |motor activity| |cellular pigmentation| |mitotic spindle organization| |kinetochore| |pigmentation| |ciliary basal body-plasma membrane docking| |antigen processing and presentation of exogenous peptide antigen via MHC class II| |antigen processing and presentation of peptide antigen via MHC class II| |antigen processing and presentation of peptide or polysaccharide antigen via MHC class II| |microtubule cytoskeleton organization involved in mitosis| |growth cone| |G2/M transition of mitotic cell cycle| |cell cycle G2/M phase transition| |vesicle| |spindle organization| |organelle localization by membrane tethering| |establishment of vesicle localization| |antigen processing and presentation of exogenous peptide antigen| |membrane docking| |antigen processing and presentation of exogenous antigen| |vesicle localization| |antigen processing and presentation of peptide antigen| |regulation of G2/M transition of mitotic cell cycle| |regulation of cell cycle G2/M phase transition| |endoplasmic reticulum to Golgi vesicle-mediated transport| |antigen processing and presentation| |mitotic cell cycle phase transition| |cell cycle phase transition| |microtubule| |cilium assembly| |establishment of organelle localization| |cilium organization| |Golgi vesicle transport| |regulation of mitotic cell cycle phase transition| |plasma membrane bounded cell projection assembly| |regulation of cell cycle phase transition| |cell projection assembly| |protein kinase binding| |microtubule cytoskeleton organization| |centrosome| |cell population proliferation| |organelle localization| |mitotic cell cycle process| |regulation of mitotic cell cycle| |microtubule-based process| |mitotic cell cycle| |protein localization to organelle| |regulation of cell cycle process| |organelle assembly| |cell cycle process| |identical protein binding| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |cell projection organization| |regulation of cell cycle| |cell cycle| |intracellular transport| |cellular protein localization| |cellular macromolecule localization| |establishment of localization in cell| |vesicle-mediated transport| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |BI-2536 0.0042μM R03 exp95|-2.81| |Fluvastatin 2.2μM R06 exp282|-1.81| |Thioridazine 5μM R08 exp530|-1.8| |BI-6727 0.0125μM R03 exp97|-1.79| |Vincristine 0.0005μM R04 exp190|-1.77| |A-395N 10μM R05 exp219|1.72| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |RRM1|0.467| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 175/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|0/1| |bile duct|4/28| |blood|9/28| |bone|5/26| |breast|14/33| |central nervous system|19/56| |cervix|0/4| |colorectal|4/17| |esophagus|3/13| |fibroblast|0/1| |gastric|1/16| |kidney|2/21| |liver|7/20| |lung|16/75| |lymphocyte|3/16| |ovary|6/26| |pancreas|7/24| |peripheral nervous system|4/16| |plasma cell|2/15| |prostate|0/1| |skin|6/24| |soft tissue|2/9| |thyroid|0/2| |upper aerodigestive|6/22| |urinary tract|5/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 786 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.92 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='DCTN2 Expression in NALM6 Cells: 5.92'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
