The encoded protein hydrolyzes DNA, is not inhibited by actin, and mediates the breakdown of DNA during apoptosis. Mutations in this gene are a cause of systemic lupus erythematosus-16. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Feb 2012]. * **<color #00a2e8>UniProt Summary</color>**: Has DNA hydrolytic activity. Is capable of both single- and double-stranded DNA cleavage, producing DNA fragments with 3'-OH ends (By similarity). Can cleave chromatin to nucleosomal units and cleaves nucleosomal and liposome-coated DNA (PubMed:9070308, PubMed:9714828, PubMed:14646506, PubMed:10807908, PubMed:27293190). Acts in internucleosomal DNA fragmentation (INDF) during apoptosis and necrosis (PubMed:23229555, PubMed:24312463). The role in apoptosis includes myogenic and neuronal differentiation, and BCR-mediated clonal deletion of self-reactive B cells (By similarity). Is active on chromatin in apoptotic cell-derived membrane-coated microparticles and thus suppresses anti-DNA autoimmunity (PubMed:27293190). Together with DNASE1, plays a key role in degrading neutrophil extracellular traps (NETs) (By similarity). NETs are mainly composed of DNA fibers and are released by neutrophils to bind pathogens during inflammation (By similarity). Degradation of intravascular NETs by DNASE1 and DNASE1L3 is required to prevent formation of clots that obstruct blood vessels and cause organ damage following inflammation (By similarity). {ECO:0000250|UniProtKB:O55070, ECO:0000250|UniProtKB:O89107, ECO:0000269|PubMed:10807908, ECO:0000269|PubMed:14646506, ECO:0000269|PubMed:23229555, ECO:0000269|PubMed:24312463, ECO:0000269|PubMed:27293190, ECO:0000269|PubMed:9070308, ECO:0000269|PubMed:9714828}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Exo endo phos| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |deoxyribonuclease I activity| |regulation of neutrophil mediated cytotoxicity| |deoxyribonuclease activity| |programmed cell death involved in cell development| |apoptotic DNA fragmentation| |DNA catabolic process, endonucleolytic| |apoptotic nuclear changes| |cellular component disassembly involved in execution phase of apoptosis| |DNA catabolic process| |regulation of acute inflammatory response| |execution phase of apoptosis| |regulation of myeloid leukocyte mediated immunity| |regulation of leukocyte mediated cytotoxicity| |regulation of cell killing| |regulation of leukocyte mediated immunity| |nucleic acid phosphodiester bond hydrolysis| |regulation of inflammatory response| |nucleobase-containing compound catabolic process| |cellular component disassembly| |heterocycle catabolic process| |cellular nitrogen compound catabolic process| |aromatic compound catabolic process| |regulation of immune effector process| |organic cyclic compound catabolic process| |neutrophil activation involved in immune response| |neutrophil activation| |granulocyte activation| |myeloid cell activation involved in immune response| |myeloid leukocyte activation| |leukocyte activation involved in immune response| |cell activation involved in immune response| |calcium ion binding| |DNA metabolic process| |regulation of defense response| |cellular macromolecule catabolic process| |apoptotic process| |leukocyte activation| |endoplasmic reticulum| |macromolecule catabolic process| |programmed cell death| |cell activation| |cell death| |immune effector process| |regulation of response to external stimulus| |DNA binding| |regulation of response to stress| |cell development| |regulation of immune system process| |organic substance catabolic process| |cellular catabolic process| |immune response| |extracellular region| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |UM0125461 0.74μM R02 exp84|-1.88| |AICAR 240μM R03 exp116|-1.81| |CB-839 10μM R05 exp224|-1.74| |VER-155008 3.9μM R06 exp301|1.99| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5785 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 1.69 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='DNASE1L3 Expression in NALM6 Cells: 1.69'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
