The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2:GTP:methionyl-tRNAi and eIF-5 to form the 43S pre-initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation (PubMed:18599441, PubMed:25849773). The eIF-3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed:25849773). In the eIF-3 complex, EIF3D specifically recognizes and binds the 7-methylguanosine cap of a subset of mRNAs (PubMed:27462815). {ECO:0000269|PubMed:18599441, ECO:0000269|PubMed:25849773, ECO:0000269|PubMed:27462815}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |eIF-3 zeta| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |cap-dependent translational initiation| |mRNA cap binding| |viral translational termination-reinitiation| |eukaryotic translation initiation factor 3 complex, eIF3m| |positive regulation of mRNA binding| |positive regulation of RNA binding| |regulation of mRNA binding| |IRES-dependent viral translational initiation| |regulation of RNA binding| |viral translation| |eukaryotic 48S preinitiation complex| |eukaryotic translation initiation factor 3 complex| |formation of cytoplasmic translation initiation complex| |eukaryotic 43S preinitiation complex| |cytoplasmic translational initiation| |translation initiation factor activity| |cytoplasmic translation| |positive regulation of translation| |viral gene expression| |translational initiation| |positive regulation of cellular amide metabolic process| |positive regulation of binding| |ribonucleoprotein complex assembly| |ribonucleoprotein complex subunit organization| |regulation of translation| |regulation of binding| |translation| |regulation of cellular amide metabolic process| |peptide biosynthetic process| |ribonucleoprotein complex biogenesis| |posttranscriptional regulation of gene expression| |amide biosynthetic process| |peptide metabolic process| |viral process| |symbiotic process| |cellular amide metabolic process| |interspecies interaction between organisms| |cellular protein-containing complex assembly| |RNA binding| |organonitrogen compound biosynthetic process| |protein-containing complex assembly| |positive regulation of cellular protein metabolic process| |cellular nitrogen compound biosynthetic process| |positive regulation of protein metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |positive regulation of molecular function| |protein-containing complex subunit organization| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |membrane| |gene expression| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Nocodazole 0.2μM R00 exp24|-1.78| |SGC2096 2.6μM R03 exp153|1.7| |Taxol 0.002μM R01 exp54|1.71| |A-366 10μM R03 exp115|1.74| |Quinacrine 2.5μM R03 exp146|1.98| |L-74142 5μM R03 exp131|1.99| |MS023 7μM R03 exp135|2.01| |THZ531 0.225μM R08 exp444|2.01| |A-395 10μM R05 exp218|2.05| |Asunaprenir 3μM R07 exp336|2.15| |Caffeine 2600μM R08 exp463|2.19| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |HGC6.3|0.403| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 667/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|26/28| |blood|26/28| |bone|24/26| |breast|31/33| |central nervous system|51/56| |cervix|3/4| |colorectal|17/17| |esophagus|12/13| |fibroblast|1/1| |gastric|14/16| |kidney|15/21| |liver|18/20| |lung|66/75| |lymphocyte|16/16| |ovary|24/26| |pancreas|20/24| |peripheral nervous system|15/16| |plasma cell|15/15| |prostate|1/1| |skin|19/24| |soft tissue|8/9| |thyroid|2/2| |upper aerodigestive|21/22| |urinary tract|26/29| |uterus|4/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1253 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.76 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='EIF3D Expression in NALM6 Cells: 7.76'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
