This protein contains an actin filament-binding domain, which together with its Dbl homology domain and one of its pleckstrin homology domains, can form microspikes. This protein can activate MAPK8 independently of the actin filament-binding domain, and it is also involved in the activation of CDC42 via the exchange of bound GDP for free GTP. The activation of CDC42 also enables this protein to play a role in mediating the cellular invasion of Cryptosporidium parvum, an intracellular parasite that infects the gastrointestinal tract. Mutations in this gene can cause Charcot-Marie-Tooth disease type 4H (CMT4H), a disorder of the peripheral nervous system. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2015]. * **<color #00a2e8>UniProt Summary</color>**: Activates CDC42, a member of the Ras-like family of Rho- and Rac proteins, by exchanging bound GDP for free GTP. Plays a role in regulating the actin cytoskeleton and cell shape. Activates MAPK8 (By similarity). {ECO:0000250, ECO:0000269|PubMed:15133042}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |RhoGEF| |PH| |FYVE| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |filopodium assembly| |small GTPase binding| |Rho guanyl-nucleotide exchange factor activity| |filopodium| |ruffle| |guanyl-nucleotide exchange factor activity| |regulation of Rho protein signal transduction| |regulation of cell shape| |lamellipodium| |regulation of Ras protein signal transduction| |actin binding| |regulation of small GTPase mediated signal transduction| |cytoskeleton| |plasma membrane bounded cell projection assembly| |cell projection assembly| |regulation of GTPase activity| |regulation of cell morphogenesis| |actin cytoskeleton organization| |actin filament-based process| |positive regulation of apoptotic process| |positive regulation of programmed cell death| |positive regulation of cell death| |Golgi apparatus| |regulation of anatomical structure morphogenesis| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |cell projection organization| |regulation of hydrolase activity| |G protein-coupled receptor signaling pathway| |regulation of apoptotic process| |regulation of programmed cell death| |regulation of cell death| |regulation of intracellular signal transduction| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Mubritinib 0.2μM R01 exp48|-1.76| |MSC2530818 10μM R07 exp379|-1.71| |RO-3307 4.7μM R02 exp80|1.73| |Pterostilbene 16μM R02 exp78|1.81| |Colchicine 0.009μM R05 exp215|1.83| |Nicotinamide 1000μM R03 exp101|2.21| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 13659 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 3.33 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='FGD4 Expression in NALM6 Cells: 3.33'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
