This enzyme has a central role in establishing and maintaining both cytosolic and mitochondrial folylpolyglutamate concentrations and, therefore, is essential for folate homeostasis and the survival of proliferating cells. This enzyme catalyzes the ATP-dependent addition of glutamate moieties to folate and folate derivatives. Alternative splicing results in transcript variants encoding different isoforms. [provided by RefSeq, Jan 2014]. * **<color #00a2e8>UniProt Summary</color>**: Catalyzes conversion of folates to polyglutamate derivatives allowing concentration of folate compounds in the cell and the intracellular retention of these cofactors, which are important substrates for most of the folate-dependent enzymes that are involved in one-carbon transfer reactions involved in purine, pyrimidine and amino acid synthesis. Unsubstituted reduced folates are the preferred substrates. Metabolizes methotrexate (MTX) to polyglutamates. {ECO:0000269|PubMed:8408018, ECO:0000269|PubMed:8408019, ECO:0000269|PubMed:8408021, ECO:0000269|PubMed:8662720}. <button type='default' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> No Pfam Domain information is available for this gene. </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |tetrahydrofolylpolyglutamate biosynthetic process| |tetrahydrofolylpolyglutamate synthase activity| |tetrahydrofolylpolyglutamate metabolic process| |folic acid-containing compound biosynthetic process| |pteridine-containing compound biosynthetic process| |folic acid metabolic process| |one-carbon metabolic process| |folic acid-containing compound metabolic process| |glutamate metabolic process| |pteridine-containing compound metabolic process| |cellular modified amino acid biosynthetic process| |glutamine family amino acid metabolic process| |animal organ regeneration| |water-soluble vitamin metabolic process| |dicarboxylic acid metabolic process| |vitamin metabolic process| |liver development| |hepaticobiliary system development| |coenzyme biosynthetic process| |regeneration| |cellular modified amino acid metabolic process| |alpha-amino acid metabolic process| |cofactor biosynthetic process| |coenzyme metabolic process| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |cellular amino acid metabolic process| |mitochondrial matrix| |mitochondrial inner membrane| |gland development| |cofactor metabolic process| |drug metabolic process| |amide biosynthetic process| |small molecule biosynthetic process| |brain development| |head development| |cellular amide metabolic process| |carboxylic acid metabolic process| |central nervous system development| |oxoacid metabolic process| |organic acid metabolic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |mitochondrion| |organic cyclic compound biosynthetic process| |organonitrogen compound biosynthetic process| |ATP binding| |cellular nitrogen compound biosynthetic process| |small molecule metabolic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Trimetrexate 0.03μM R08 exp535|-2.35| |Dimethyl-Sulfoxide 1pc R03 exp120|-2.29| |TNF-alpha 44ng/ml R08 exp533|1.73| |Aminopterin 0.005μM R07 exp335|1.76| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 25/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|1/28| |blood|5/28| |bone|0/26| |breast|2/33| |central nervous system|0/56| |cervix|0/4| |colorectal|1/17| |esophagus|0/13| |fibroblast|0/1| |gastric|1/16| |kidney|2/21| |liver|1/20| |lung|2/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|1/24| |peripheral nervous system|0/16| |plasma cell|1/15| |prostate|0/1| |skin|3/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|1/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 70 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.11 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='FPGS Expression in NALM6 Cells: 7.11'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
