Members of this family regulate group 1 metabotrophic glutamate receptor function. The encoded protein is a postsynaptic density scaffolding protein. Alternative splicing results in multiple transcript variants. Two related pseudogenes have been identified on chromosome 14. [provided by RefSeq, Jun 2011]. * **<color #00a2e8>UniProt Summary</color>**: Postsynaptic density scaffolding protein. Binds and cross-links cytoplasmic regions of GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1 and SHANK3. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, it aids the coupling of surface receptors to intracellular calcium release. May also couple GRM1 to PI3 kinase through its interaction with AGAP2. Isoforms can be differently regulated and may play an important role in maintaining the plasticity at glutamatergic synapses (PubMed:9808459). Required for normal hearing (PubMed:25816005). {ECO:0000269|PubMed:25816005, ECO:0000269|PubMed:9808459}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |WH1| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |G protein-coupled glutamate receptor binding| |GKAP/Homer scaffold activity| |chemical homeostasis within a tissue| |regulation of store-operated calcium entry| |negative regulation of calcineurin-NFAT signaling cascade| |negative regulation of calcineurin-mediated signaling| |G protein-coupled glutamate receptor signaling pathway| |stereocilium tip| |behavioral response to cocaine| |negative regulation of interleukin-2 production| |calcium-mediated signaling using intracellular calcium source| |negative regulation of calcium-mediated signaling| |regulation of calcineurin-NFAT signaling cascade| |regulation of calcineurin-mediated signaling| |glutamate receptor signaling pathway| |regulation of interleukin-2 production| |response to cocaine| |apical part of cell| |response to anesthetic| |regulation of calcium-mediated signaling| |response to alkaloid| |regulation of G protein-coupled receptor signaling pathway| |adult behavior| |sensory perception of sound| |calcium-mediated signaling| |sensory perception of mechanical stimulus| |response to ammonium ion| |tissue homeostasis| |postsynaptic membrane| |protein domain specific binding| |regulation of calcium ion transport| |postsynaptic density| |actin binding| |negative regulation of cytokine production| |response to xenobiotic stimulus| |neuron projection| |multicellular organismal homeostasis| |second-messenger-mediated signaling| |anatomical structure homeostasis| |glutamatergic synapse| |neuronal cell body| |regulation of metal ion transport| |dendrite| |protein heterodimerization activity| |negative regulation of intracellular signal transduction| |cell junction| |behavior| |regulation of cytokine production| |regulation of ion transport| |protein homodimerization activity| |response to organic cyclic compound| |sensory perception| |response to organonitrogen compound| |response to drug| |response to nitrogen compound| |chemical homeostasis| |negative regulation of multicellular organismal process| |negative regulation of signal transduction| |G protein-coupled receptor signaling pathway| |negative regulation of cell communication| |negative regulation of signaling| |nervous system process| |response to oxygen-containing compound| |negative regulation of response to stimulus| |homeostatic process| |positive regulation of signal transduction| |intracellular signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |regulation of transport| |system process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |BI-D1870 3.15μM R02 exp66|-1.98| |WYE-354 6μM R02 exp90|-1.85| |A-366 10μM R03 exp115|-1.81| |Ribavirin 10μM R01 exp52|-1.73| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 8777 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 4.31 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='HOMER2 Expression in NALM6 Cells: 4.31'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
